Abstract
ÖZET Bu çalışmada Pt elektrot yüzeyine kaplanan poli(vinilferrosenyum), PVF+, filmine laktat oksidaz (LOD)'ın tutuklanmasıyla oluşturulan enzim elektroduyla laktatın kararlı hal amperometrik ölçümleri yapıldı. Poli(vinilferrosen), PVF.'in 0.1 M tetrabütilamonyumperklorat (TBAP) içeren metilen klorürdeki çözeltisinin Pt electrotda 0.7 V da yapılan elektrolizi ile PVF+CI04` filmi oluşturuldu. Enzimin polimer filminde tutuklanması için LOD'ın izoelektrik noktasının (4.6) üzerindeki bir pH değeri olan pH 7.0'de 0.01 M fosfat tamponunda enzim çözeltisi hazırlandı. Bu pH değerinde negatif yüklü olan enzimin, LOD`, polimer film yapısındaki CIO4` karşıt anyonuyla yer değiştirmesiyle PVF+LOD` enzim elektrodu oluşturuldu. Polimer film yapısındaki enzimin nitel ve nicel analizinde ise UV spektroskopisinden yararlanıldı. PVF ve PVF+CI04`'ın dönüşümlü voltametrik davranışları da incelendi. Deneysel ortam için en uygun koşulların belirlemek amacıyla pH ve sıcaklık etkisi araştırıldı. En yüksek akım değerleri pH 7.0 ve 37°C de ölçüldü. Bu enzim elektrodunun doğrusal derişim aralığı 0.0525 - 1.365 mM olarak bulundu. En uygun çalışma sıcaklığını belirlemek amacıyla 30 - 42°C aralığında yapılan amperometrik ölçümlerden yararlanarak laktatın enzimatik oksidasyon tepkimesi için aktivasyon enerjisi bulundu. Polimer film yapısında bulunan LOD enziminin laktata ne kadar uyumlu olduğunu belirlemek amacıyla en uygun çalışma koşullarında elde edilen akım - derişim verilerinden yararlanılarak hem immobilize hem de serbest LOD için Lineweaver - Burk grafiği oluşturuldu ve Michealis - Menten sabiti hesaplandı. Geliştirilen enzim elektrodu ile aynı substrat derişimi için birer gün ara ile ölçülen akım değerlerinden yararlanılarak enzim elektrodunun kararlı - aktif ömrü 15 gün olarak belirlendi. ABSTRACT The steady state amperometric measurements of lactate oxidase was performed by an enzyme electrode which was developed through via immobilization of the lactate oxidase (LOD) in polyvinyl ferrocenium), PVF/ film coated on a Pt electrode. PVF+CI04` film was deposited on Pt electrode at + 0.7 V vs Ag / AgCI through oxidation of poly(vinylferrocene), PVF, in methylene chloride which contained 0.1 M tetrabuthylammoniumperchlorate (TBAP). The preparation of the enzyme electrode was accomplished by immersing the PVF+CI04` coated Pt electrodes in the enzyme solution. The pH of the enzyme solution was kept at 7 which was well above the isoelectric points of LOD (4.6). The enzyme exists in the form of anions (LOD) under these conditions and the ion exchange type interaction of the enzyme with the oxidized polymer (PVF+) occurs. UV spectroscopic studies were also done for the quantitative and qualitative analyses of the enzyme in the polymeric matrix. Cyclic voltammetric behaviors PVF and PVF+CI04` have also been studied. For determination the optimum conditions the effect of the temperature, pH and concentrations were investigated. It was found that the maximum current was obtained at 37°C, at pH 7.0 and LOD solutions that contained 0.375 mg protein mL`1. The linear concentration range was found to be between 0.0525 - 1.365 mM for this enzyme electrode. Amperometric measurements were performed at the interval 30 - 42°C for the LOD to find out the activation energy of the enzymatic oxidation reaction of lactate and also the optimum operational temperature. Lineweaver - Burk graph was plotted and apparent Michealis - Menten constant was calculated respectively for the immobilized and the free enzyme system. The stability - time of the developed enzyme electrode was found to be about 15 days using the current values measured daily for the same substrate concentration.
