Abstract
Saccharomyces cerevisiae KALECİK 1 ve Saccharomyces cerevisiae NARİNCE 3 ŞARAP MAYASI SUŞLARININ ÜREME VE GLİSERİN ÜRETİM KİNETİKLERİNİN İNCELENMESİ Seda Karasu Hacettepe Üniversitesi, Gıda Mühendisliği Bölümü ÖZ Bu araştırmada, iki şarap mayası suşu dan S. cerevisiae Kalecik 1 ve S. cerevisiae Narince 3'ün üreme ve gliserin üretim kinetikleri incelenmiştir. Çalkalamalı su banyolarında gerçekleştirilen çalışmalarda, substrat türlerinin ve başlangıç derişimlerinin etkilerinin incelenmesinde glukoz, fruktoz ve sakkaroz kullanılmıştır. Her iki suş için de glukoz, fruktoz ve sakkarozun, mayaların üremesi için gereken optimum substrat derişimlerinden daha yüksek başlangıç derişimlerinde inhibisyon etkisi görülmüştür. S. cerevisiae Kalecik 1 ve S. cerevisiae Narince 3 suşlarının üremede kullanmaya en yatkın oldukları substratlar sırasıyla, glukoz ve sakkaroz olarak bulunmuştur. Fruktozlu ortamda her iki susun da düşük üreme eğilimi gösterdikleri, ancak diğer substratlarla kıyaslandığında gliserin üretimlerinin oldukça yüksek olduğu belirlenmiştir. S usların üremesi için besiyeri başlangıç optimum pH değeri 4.00 olarak bulunmuştur. S. cerevisiae Kalecik 1'in pH 5.92'de, S. cerevisiae Narince 3'ün ise pH 6.27'de maksimum düzeyde gliserin ürettikleri belirlenmiştir. Her iki susun da 30°C'de maksimum özgül üreme hızına ulaştıkları, suşların gliserin üretimi için gerekli en uygun sıcaklık aralığının ise 25-30°C olduğu tespit edilmiştir. Gliserin üretimi için en uygun aşı derişimi her iki suş için de %7.5 (v/v) olarak bulunurken, üreme için bu değerler Kalecik 1 ve Narince 3 suşları için sırasıyla, %10 ve %7.5 olarak belirlenmiştir. 0.3 g/L başlangıç amonyum sülfat derişiminde her iki susun da maksimum düzeyde gliserin ürettikleri belirlenmiştir. S. cerevisiae Kalecik 1 ve Narince 3 için maksimum özgül üreme hızının elde edildiği başlangıç amonyum sülfat derişimleri sırasıyla, 1.0 g/L ve 0.8 g/L olarak tespit edilmiştir. Fermantasyon ortamı olarak üzüm suyu kullanıldığında Kalecik 1 ve Narince 3 suşlarının ürettikleri maksimum gliserin derişimleri ise sırasıyla, 1 1.8 g/L ve 14.1 g/L olarak belirlenmiştir. In the studies which were carried out in water bath shakers, glucose, fructose and sucrose were used to determine the effects of types and initial concentrations of substrates. For both strains, inhibition effect was observed at initial substrate concentrations of glucose, fructose and sucrose which were above the optimum substrate concentrations necessary for the yeasts growth. Substrates, which S. cerevisiae Kalecik 1 and S. cerevisiae Narince 3 showed most tendency to use for growth were found as glucose and sucrose, respectively. In the medium containing fructose, it was determined that both of the strains showed low tendency for growth, but very high glycerol production levels were obtained when compared with the other substrates. Optimum initial pH value of the medium for growth of strains was found as 4.00. S. cerevisiae Kalecik 1 was found to produce glycerol at maximum levels at pH 5.92, and S. cerevisiae Narince 3 at pH 6.27. It was determined that, both strains have reached maximum specific growth rates at 30°C, while optimum temperature range for production of glycerol was found as 25-30°C. The most suitable inoculation concentration for glycerol production of both strains was found as %7.5 (v/v), while these values were %10 and %7.5 for the growth of Kalecik 1 and Narince 3 strains, respectively. At an initial ammonium sulphate concentration of 0.3 g/L, both of the strains produced glycerol at maximum levels. The initial ammonium sulphate concentrations at which maximum specific growth rates were obtained for S. cerevisiae Kalecik 1 and Narince 3 were determined as 1.0 g/L and 0.8 g/L, respectively. When grape juice was used as fermentation medium, maximum glycerol concentrations produced by Kalecik 1 and Narince 3 strains were found as 1 1.8 g/L and 14.1 g/L, respectively.
