Tuz stresinde büyütülen ayçiçeği bitkilerinde zamana bağlı olarak serbest ve bağlı poliamin düzeylerinin değişimi ve bunun bazı fizyolojik parametrelerle ilişkisi

Abstract
Bu çalışmada ayçiçeği (Helianthus annuus L)'nin tuza dayanıklı (Çoban) ve duyarlı (Sanbro) olan çeşitlerinde tohum, kök, gövde ve yaprak dokularında serbest, bağlı ve total poliamin düzeyleri, ayrıca yaprak dokularında prolin, protein ve bazı iyon (Na+, K*, Cl`, K7Na+) miktarları üzerine değişik konsantrasyonlardaki tuz stresinin (50, 100 ve 150mM NaCI) etkileri zamana bağlı olarak (5, 15 ve 25. gün) araştırılmıştır. Kuru tohumlarda serbest Spm., Spd., AS-bağlı Spd. ve total poliamin düzeylerinin tuza duyarlı çeşit olan Sanbro'da, dayanıklı olan Çoban çeşidine göre daha yüksek olduğu tespit edilmiştir. Yirmidört saat süreyle değişik konsantrasyonlardaki tuzlu kültür çözeltilerinde ıslatılmış tohumlarda tuz stresi, Sanbro tohumlarında serbest ve AS-bağlı Spm. düzeylerinde, ayrıca her iki çeşitte de serbest, AS-bağlı, AINS-bağlı ve total Spd., AINS-bağlı ve total Spm. ve düzeylerinde azalmaya neden olmuştur. Serbest Put. ise kuru tohumlarda tespit edilememiş, tuzlu kültür çözeltileri ile ıslatılan tohumlarda eser düzeyde tespit edilebilmiştir. Değişik sürelerde tuz stresine maruz kalan ayçiçeği bitkilerinin farklı dokularında (kök, gövde ve yaprak) serbest, AS-bağlı ve total Spm. miktarı artmış, fakat diğer poliamin çeşitlerinin düzeyinde genellikle ya azalma görülmüş ya da herhangi bir değişiklik saptanamamıştır. Tuz uygulaması ile yaprak dokularındaki prolin, protein ve çeşitli iyon (Na+, K/ Cl`, ve K7Na+) düzeyleri de tuz konsantrasyonu, zaman ve çeşide bağlı olarak genel anlamda önemli değişimler göstermiştir. Çalışmanın esas amacı, tuz stresi altındaki bitkilerde poliamin birikiminin, strese adaptasyon mekanizmasında ve tuza dayanıklı çeşitlerin seçilmesinde birer kriter olarak kullanılıp kullanılamayacağının belirlenmesidir. Bu nedenle tuz-poliamin ilişkisi ve incelenen diğer fizyolojik parametreler arasındaki muhtemel bağlantılar ayrıntılı bir şekilde tartışılmıştır. In the present study, the effect of salt stress (50, 100, 150mM NaCI) on the endogenous levels of free, bound and total polyamines was studied in seeds, root, shoot and leaf tissues of salt-tolerant (Coban) and salt-sensitive (Sanbro) cultivars of sunflower (Helianthus annuus L) plants for 5, 15 and 25 days of the growth period. The amounts of proline, protein and some ions (Na+, K/ CI`, K7Na+) in the leaf tissues of the cultivars were also determined. Levels of free Spm., Spd., AS-bound Spd. and total polyamine were found to be higher in dry seeds of salt-sensitive Sanbro than salt-tolerant Coban cultivar. Soaking the seeds, for 24 hours, in different concentrations of saline culture solutions, caused to decrease the free and AS-bound Spm. levels only in Sanbro but decreased the free, AS-bound, AINS-bound and total Spd. with AINS-bound and total Spm. levels in both cultivars. The free Put. could not be detected in dry seeds whereas a trace amount of Put. was found in the soaked seeds. The amounts of free, AS-bound and total Spm. were increased in different tissues (roots, shoots and leaves) of sunflower plants subjected to salt stress for different periods of time while the levels of other polyamine titers were decreased in general or any significant change could not be determined. The levels of proline, protein and various ions (Na+, K*, CI`, K7Na+) in the leaf tissue have changed depending on salt concentration, period of time and variety of the plant used. The main target of the study is to determine if any kind of polyamine accumulating capacity of the plants under the salt stress, could be used as a criterium for the adaptation mechanism and for the selection of the salt-tolerant variety. Therefore, possible links between the salt-polyamine relation and the other physiological parameters studied were discussed in detail.
