Abstract
BROİLERLERDEN KLASİK KÜLTÜREL YÖNTEMLERLE SALMONELLA İZOLASYONUNDA ÇEŞİTLİ YÖNTEM MODİFİKASYONLARININ DENENMESİ Betül Vazgeçer ÖZ Bu çalışmada; klasik kültürel yöntemlerin çeşitli işlem aşamalarında modifikasyonlar yaparak broilerlerden Salmonella izolasyonunda yöntem duyarlılığının artırılması ve süresinin kısaltması amaçlanmıştır. Çalışmanın ilk aşamasında; Türk Standartları Enstitüsü'nün TS 7438 nolu standardındaki klasik kültürel yöntem kullanılarak broiler deri örneklerindeki Salmonella ve beraberindeki rekabetçi flora üyeleri belirlenmiştir. API 10 S sistemi ile broilerlerden izole edilen toplam 155 adet izolattan 11 adedi Salmonella spp. olarak tanımlanmıştır. API 10 S sistemi ile Salmonella spp. olarak tanımlanan bazı izolatların API 20 E sistemi kullanılarak gerçekleştirilen testler sonucunda Hafnia alvei (3 adet), Citrobacter freundii (2 adet), Cit. braakii (1 adet), Enterobacter aerogenes (1 adet) ve Ent. cloacae (1 adet) ve 3 adedinin de Salmonella olduğu belirlenmiştir. Rekabetçi flora üyeleri arasından seçilen 61 adet bakteri izolatının novobiyosin (105 ug/mL), tergitol-4 (39 ul/mL) ve eritromisin (30 ug/mL)'e duyarlılık oranları sırasıyla % 79, % 49 ve % 33 olarak bulunmuştur, önzenginleştirme besiyeri olarak 105 ug/mL novobiyosin eklenmiş Universal Enrichment Broth (UB) besiyeri ve zenginleştirme besiyeri olarak da Rappaport-Vassiliadis (RV) broth besiyerinin kullanıldığı model sistemde; selektif katı besiyerlerinde gelişen Salmonella kolonileri ile rekabetçi floraya ait kolonilerin birbirlerinden en iyi şekilde ayrıldığı sonucuna varılmıştır (p<0,05). Önzenginleştirme besiyeri olarak novobiyosin eklenmiş UB besiyeri ve zenginleştirme besiyeri olarak RV broth besiyerinin kullanıldığı ve bu besiyerinin de 6 saat inkübe edilmesi şeklindeki yöntem modifikasyonunun broilerden Salmonella izolasyonunda klasik kültürel yönteme göre daha iyi sonuç verdiği belirlenmiştir. Böylece broilerlerden Salmonella izolasyonunda toplam test süresinin 18 saat kısaltılması sağlanabilmiştir. In the first phase of the study; Salmonella and competitive bacteria of broiler breast skin samples were determined using the standard cultural method as described in the standard of Turkish Standard Organization with the number TS 7438. Totally 11 of 155 isolates from broiler were identified as Salmonella spp. using API 10 S identification system. Isolates identified as Salmonella spp. by API 10 S system were then identified as Hafnia alvei (3), Citrobacter freundii (2), Cit. braakii (1), Enterobacter aerogenes (1) and Ent. cloacae (1) and also Salmonella (3) using API 20 E identification system. The susceptibility rates of 61 isolates chosen from competitive bacteria to novobiocin (105 ug/mL), tergitol-4 (39 ul/mL) and erythromycine (30 ug/mL) were found as 79%, 49% and 33%, respectively. It was concluded that the colonies of Salmonella and competitive bacteria on selective plating media were well differentiated by the model system including Universal Enrichment Broth (UB) with 105 ug/mL novobiocin as pre-enrichment broth and subsequently Rappaport-Vassiliadis (RV) broth as enrichment broth (p<0.05). The modified method including UB with novobiocin as pre-enrichment broth and subsequently RV broth as enrichment broth with the incubation time of 6 hours gave better results than standard cultural methods for the isolation of Salmonella from broiler on selective plating media. Hence, it has been achieved that the total test time of the isolation of Salmonella from broiler could be reduced to 18 hours.
