Abstract
Sunulan çalışmanın amacı, doğal bir polimer olan kitosandan hazırlanan membranyüzeylerinde biyofilm oluşum mekanizmasının ve kitosan membranların bakteriyalenzim ile bozunma kinetiğinin incelenerek, kitosanın biyomalzeme olarakkullanımının araştırılmasıdır.Yapılan araştırmanın ilk kısmında, gözeneklilik ve çapraz bağlanma açısındanfarklılık gösteren kitosan membranlar üretilmiştir. Gözenekli membranlar, kitosançözeltisinin silika partikülleri ile homojen olarak karıştırılması ve sonrasında bupartiküllerin ortamdan uzaklaştırılmasıyla hazırlanmıştır. Zayıf asitlerde çözünenkitosan membranlarda, çözünmeyi engellemek amacıyla, epiklorohidrin kullanılarakçapraz bağlanma sağlanmıştır. Böylelikle, farklı özelliklere sahip kitosanmembranlarda, bakteriyal yapışma ve biyofilm oluşumunun incelenmesi mümkünolmuştur. Kitosan, bakteriler tarafından üretilen kitosanaz ile degradeedilebilmektedir. Çalışmada kullanılan B.subtilis ve P.aeruginosa kitosanazenzimine sahiptirler. Çalışma sırasında, degradasyon kinetiği, Streptomycesgriseus'den izole edilmiş kitosanaz ile asetat tamponunda gerçekleştirilmiştir.Hazırlanan kitosan membranlarda bakteriyal yapışma kinetiği incelenmiştir. Buçalışmada, biyomalzeme enfeksiyonlarında önemli rol oynayan Escherischia coli,Pseudomonas aeruginosa, Streptococcus epidermidis ve Bacillus subtiliskullanılmıştır. Bakteriyal yapışma kinetiği, karşılaştırmalı olarak, UV-Spektrofotometre, Koloni oluşturma birimi ve ATP (Adenozin trifosfat)Biyoluminesans yöntemleri ile incelenmiştir. P.aeruginosa, en fazla yapışmagösteren tür olmuştur. Bunu sırasıyla B.subtilis, S.epidermidis ve E.coli izlemiştir.Biyomalzemelerde biyofilm oluşumu, yukarıda belirtilen mikroorganizmalar ileyürütülmüştür. Yapışma sonuçları ile uyumlu olarak, kitosan membranlarda en çokiP.aeruginosa üremiştir, daha sonra sırasıyla B.subtilis, S.epidermidis ve E.coligelmiştir. Üreyen mikroorganizma sayısı, ATP biyoluminesan, biyofilm oluşumu iseSEM ve konfokal mikroskopi ile incelenmiştir. Üreyen mikroorganizmaların biyofilmoluşturmadıkları görülmüştür.Kitosan membranlar gözeneksiz ve iki farklı gözenek çapına sahip olarakhazırlanmıştır. Yapılan incelemelerde gözenekli membranlarda bakteriyal yapışmave üremenin artış gösterdiği görülmüştür. Gözenek çapı büyük olan membranda budeğerlerin en yüksek olduğu görülmüştür. Kitosan membranlar, çapraz bağsız veepiklorohidrin çapraz bağlı olarak hazırlanmıştır. Kitosanaz üreten B.subtilis veP.aeruginosa, kitosan membranı degrade ettiğinden dolayı, kitosanaz üretmeyenE.coli ve S.epidermidis'den daha fazla ürediği görülmüştür.Anahtar Kelimeler: Mikrobiyal yapışma, Kitosan, Biyofilm, ATP Biyoluminesans,DegradasyonDanışman: Prof. Dr. Menemşe Gümüşderelioğlu, Hacettepe Üniversitesi, KimyaMühendisliği / Biyomühendislik Anabilim Dalıii The purpose of this study is to investigate the usage of chitosan as a biomaterialby biofilm formation mechanism on the prepared natural polymer chitosanmembranes and also by bacterial enzymatic degradation of them.In the first part of the study, chitosan membranes were produced. They differ byporosity and cross-linker. Porous chitosan membranes were prepared by stirringsilica particulars homogenously and after that, remove them from the membranes.To avoid chitosan membrane solution, which occurs in weak acids, cross-linkagewith Epichlorohydrine was performed. In this way, it was possible to investigatebacterial attachment and biofilm formation on different chitosan membranes.Chitosan, can be degradated by bacteria which produce chitosanase. B.subtilisand P.aeruginosa produce chitosanase. The degradation kinetics was done inacetate tampon by the chitosanase isolated from Streptomyces griseus.Bacterial attachment kinetics was studied on the prepared chitosan membranes.Escherischia coli, Pseudomonas aeruginosa, Streptococcus epidermidis andBacillus subtilis, that takes important role in biomaterial infections, were used forthis study. P.aeruginosa showed the most attachment. This was followed byB.subtilis, S.epidermidis and E.coli. Bacterial attachment kinetics was investigatedcomparatively by UV-Spectrophotometer, colony forming unit and ATP (adenosinetriphosfate) Bioluminescence.Biofilm formation on biomaterials was carried out with the above microorganism.P.aeruginosa showed the most proliferation. B.subtilis, S.epidermidis and E.colifollowed in order that was harmonious with the attachment study results. Theproliferated cell count was investigated with ATP Bioluminescence; biofilmformation was investigated with scanning electron microscope (SEM) and confocaliiimicroscopy. It was observed that no biofilm was formed by these proliferatedmicroorganisms.Chitosan membranes were prepared as non-porous and porous with two differentpore diameters. It was observed that bacterial attachment and proliferationincreased at porous membranes. The highest quantity was at the membranes withthe larger pore diameter. Chitosan membranes were prepared without cross-linkingand cross-linked with Epichlorohydrine.
