Bal şarabı üretimi için maya seçimi ve fermantasyon koşullarının optimizasyonu

Abstract
Bu çalışmada, ülkemizde üretimi ve tüketimi hala yapılmamakta olan ?Bal şarabı? adıverilen yeni bir ürün için teknik bilgi geliştirmek amaçlanmıştır. Fermantasyonçalışmalarında Ayçiçek (Helianthus annuus sp.) balı kullanılmıştır. Bal şarabıüretiminde ticari şarap mayaları ile farklı ballarından izole edilmiş yabani mayalarkullanılmıştır. Fermantasyonda ortamdaki gaz (CO2) oluşumu ve etil alkol oluşumukantitatif olarak izlenmiştir. Mayalar tarafından tüketilen toplam şeker miktarı Anthronemetodu ile tayin edilmiştir. Oluşan etil alkol miktarı ise Hidrometre, Vinometre ve GC-MS metotları kullanılarak karşılaştırmalı olarak tayin edilmiştir. Fermantasyondaoluşan bir yan ürün olan gliserol miktarı ise Boehringer-Mannheim enzimatik kitleri ileölçülmüştür. En yüksek gliserol üretimi 5,597 g/L olarak Pamuk balı izolatındagözlenmiştir. Fermantasyonda şekeri fermente etme gücü (%89), fermantasyonsonunda oluşturduğu alkol miktarı %12(v/v) ve fermantasyon süresi (15.saatte ilk CO2cıkışı) bakımından ticari bir şarap mayası olan Gervin Wine Yeast Varietal BSaccharomyces cerevisiae Narbonne Selection(GVY VB) potent maya suşu olarakseçilmiştir.Daha sonraki aşamada fermantasyonu kontrol eden optimize koşullarınsaptanması için çalışmalar yapılmıştır. Optimum fermantasyon koşulları olarakfermantasyon sıcaklığı: 300C, pH:3.5, şıra içeriğindeki bal miktarı 0.37kg bal/L, inoküleedilen maya miktarı 20ml, ortama ilave edilen maya nutrienti miktarı 0,65g/L olaraksaptanmıştır.Çalışmanın son aşamasında ise saptanan bu optimum koşullarda potentsuş ile gerçekleştirilen fermantasyonlarda üretilen maksimum etil alkol miktarı%12.5(v/v) ( Fermantasyonun1. günü sonunda üretilen etil alkol miktarı ise %3.5)Fermantasyonda şekeri fermente etme gücü %90.2 ve gliserol miktarı ise 3.96g/Lolarak hesaplanmıştır.Anahtar Kelimeler: Bal şarabı, Bal, Şarap, Fermantasyon, Maya, OptimizasyonDanışman: Prof.Dr. Nazif KOLANKAYA, Hacettepe Üniversitesi, Biyoloji Bölümü,Biyoteknoloji Anabilim DalıI In this study, it was aimed to improve technical information for a new product called?Honey wine? which is not yet produced and consumed in Turkey. Sunflower Honey(Helianthus annuus sp.) was used in the fermentation studies. Commercial wineyeasts and yeasts isolated from different honey types were used for the production ofHoney wine(Mead). During the fermentation process the forming of CO2 gas andethanol formation was examined quantitatively. Total sugar content consumed by theyeast was measured with Anthrone method. Ethanol content was measured both byhydrometer, vinometer and GC-MS methods comparatively. Glycerol, which is a byproduct of fermentation process, content was measured with Boehringer-Mannheimenzymatic kits. Maximum glycerol production was observed as 5,597 g/L from CottonHoney isolate yeast. Gervin Wine Yeast Varietal B Saccharomyces cerevisiaeNarbonne Selection (GVY VB) yeast was selected as a potent yeast strain becauseof its ability for the utilization of sugar (89% g/Lconsumption), the content of alcoholproduced at the end of the fermentation as 12% (v/v) and its fermentation rate ( CO2observation start time 15.ist hour).In the next step studies were done in order todetermine the optimize conditions controlling the fermentation process. Fermentationtemperature: 300C, pH= 3.5, the amount of honey in Must: 0.37kg bal/1L, the amountof yeast inoculated: 20 ml and amount of yeast nutrient added to the Must: 0.65 g/Lwere determined as the optimum fermentation conditions. In the last step of thisstudy, by using the potent yeast strain; maximum ethanol content produced by thepotent yeast strain was measured as 12.5% (v/v) ( The content of ethanol produced atthe end of the first day of fermentation is measured as 3.5%), the capacity of utilizingsugar as 90.2% and amount of glycerol as 3.96g/L in the optimum determinedfermentation conditions.IIKey words: Mead, Honey, Wine, Fermentation, Yeast, OptimisationAdvisor: Prof.Dr. Nazif KOLANKAYA, Hacettepe University, Department of Biology,Biotechnology SectionIII
