Abstract
Poecilimon similis similis'in (Retowski, 1889) (Yeşil Çekirge, Tettigoniidae: Orthoptera) bakteriyal florası ve entomopatojenlerinin belirlenerek bu zararlıya karşı mücadele etmeninin araştırması amaçlı yapılan bu çalışmada, ilk olarak zararlıdan 12 adet bakteriyal izolat ve 1 adet fungal izolat elde edildi. Bu izolatlardan bakteriyal izolatların morfolojik, fizyolojik, biyokimyasal ve moleküler özellikleri, fungusun ise sadece moleküler özellikleri belirlendi.Bakteriyal flora üyelerinin zararlı üzerindeki insektisidal etkileri araştırıldı. PSS11 olarak kodlandırılan ve Serratia marcescens olduğu saptanan izolatın 5. günde zararlı üzerinde %100'lük ölüm etkisine sahip olduğu tespit edildi. PSS5 olarak kodlandırılan izolatın ise yapılan kimyasal testler ve 16S rRNA analiz sonuçlarının NCBI genom veri tabanında bulunmaması nedeni ile yeni tür olma ihtimali bulunmaktadır.Sonraki çalışmalarda bu izolatın uygun bir formülasyonda Poecilimon similis similis'in ve benzer Poecilimon ve Isophya cinslerindeki zararlı yeşilçekirgelere karşı uygulanması planlanmaktadır. The purpose of this study is to investigate the fight against the pest Poecilimon similis similis (Retowski, 1889) (Green Grasshopper, Tettigoniidae: Orthoptera) by determining it's bacterial flora and entomopathogens. At first, twelve bacterial isolate and one fungal isolate were obtained from the pest. The morphological, physiological, biochemical and molecular features of the bacretial isolates and molecular feature of the fungal isolate were determined.The insecticidal effects of the members of bacterial flora on the pest were investigated. It is found that Serratia marcescens isolate which was coded as PSS11 have a 100 % fetal effect on the pest within 5 days. The isolate PSS5 is likely to be a new species because the results of biochemical tests and 16S rRNA analysis of the isolate could not be found on NCBI genome database.This isolate can be used to control the harmful green grasshoppers which could be found on Poecilimon similis similis and Pocelimon and Isophya species.
