Abstract
Nozokomikal enfeksiyon etkeni olarak Metisilin Dirençli Staphylococcus aureus(Methicillin Resistant Staphylococcus aureus-MRSA) görülme sıklığındaki artış vevankomisine dirençliliğin oluşması izolat karakterizasyonunun hızlı ve güvenilir birşekilde yapılmasını ve hastanelerde klonal yayılımın tanımlanmasınıgerektirmektedir.Bu çalışmanın amacı, klinik izolatlardan elde edilen Staphylococcus aureus suşlarınaait RAPD bant modellerindeki varyasyonu ve bunlar arasındaki sistematik ilişkiyiortaya çıkarmaktır. Klinik izolatlara disk difüzyon metodu kullanılarak antibiyotikduyarlılıklarına bakıldı. Bütün izolatlar vankomisine duyarlı bulundu. Bu çalışmada;MSSA için 6 (iki baskın tip-%29.4) ve MRSA için 8 (bir baskın tip-%26.6) farklıantibiyotip paterni belirlendi. 47 S.aureus ve kontrol olarak S.aures ATCC 29213,S.epidermidis ve S.saprophyticus 25 farklı primer ile rastgele çoğaltılmış polimorfikDNA (RAPD) PCR tekniği kullanılarak analiz edildi. Tüm veriler ile UPGMA metodukullanılarak dendogram çizildi. Rastgele seçilmiş 25 primerden 15'i informatifbulundu. Buna ek olarak; RAPD PCR metodu ile izolatlar MRSA ve MSSA olmaküzere ayrılmıştır. İzolatlardaki genetik uzaklık 0.1142'den 0.9565'ye değişenaralıktadır. Çalışmadan elde edilen veriler bize izolatların antibiyotip paternleri ileRAPD paternlerinin birbiri ile benzer olduğunu göstermektedir.Anahtar Kelimeler: Staphylococcus aureus, RAPD-PCR, UPGMA. The increase in the frequency of Methicillin Resistant Staphylococcus aureus(MRSA) as the causal agent of nosocomial infection and the possibility of emergenceof resistance to vancomycin demands a quick and trustworthy characterization ofisolates and identification of clonal spread within hospitals.The objective of this study is to establish variations of RAPD band patterns inStaphylococcus aureus strains which are collected from clinical isolates and itssystematic relationship between them. Antimicrobial susceptibility tests of clinicalisolates were performed by the disc diffusion method. All the isolates weresusceptible to vancomycin. In this study different antibiotype patterns weredetermined; 6 for MSSA (with two dominant patterns-%29.4) and 8 for MRSA (withone dominant pattern-%26.6). Randomly amplified polymorphic DNA (RAPD) PCRmethod with 25 different primers was analysed on 47 clinical isolates ofStaphylococcus aureus and S.aures ATCC 29213, S.epidermidis andS.saprophyticus as controls. All the data were employed to construct dendrogramsusing the UPGMA. Randomly selected 15 primers were informative out of 25. Inaddition, RAPD PCR method differentiated between MRSA and non-MRSA based onthe clustering.The overall genetic distances ranged from 0.1142 to 0.9565 among theisolates. These data suggested that the antibiotype patterns of the isolates wereidentical to RAPD patterns.Key Words: Staphylococcus aureus, RAPD-PCR, UPGMA.
