Abstract
Bitki gen kaynaklarının etkin bir sekilde kullanımı ve korunması amacıyla genetikçesitliligin karakterizasyonuna ihtiyaç vardır. Bitki genomlarını karakterize etme vetanımlamada kullanılan çesitli markır sistemleri bulunmaktadır. Bu sistemlerdenbiri olan RAPD-PCR, bugday ve diger bitki türlerinde genetik çesitliliginsaptanmasında kullanılan etkin bir tekniktir.Bu çalısmanın amacı, Türkiye'de yetisen yabani emmer ve gernik (kültüre alınmısemmer) bugdaylarının RAPD-PCR teknigi ile analiz edilerek, aralarındaki genetikçesitliligin moleküler düzeyde saptanmasıdır. Bu amaçla 11 adet yabani emmer ve8 adet gernik bugdayı RAPD-PCR teknigi kullanılarak analiz edilmistir. Buçalısmada kullanılan 25 adet primerden 20 tanesinin informatif oldugubelirlenmistir. RAPD primerlerinden elde edilen toplam 178 PCR ürününden 85'ipolimorfik bulunmustur. Çalısılan yabani emmer ve gernik bugdayı örneklerindepolimorfizm oranı %47.75 olarak saptanmıstır. Bütün veriler Aritmetik OrtalamayıKullanan Agırlıksız Çift Grup Metodu ile (UPGMA) degerlendirilmis ve fenogrambu yöntemle çizilmistir. UPGMA analizi, TUR 02456 kayıt numaralı gernik bugdayıile TUR 03399 kayıt numaralı yabani emmer örneginin birbirine en uzak; TUR03562 ve TUR 03564 kayıt numaralı gernik bugdayı örneklerinin birbirine en yakınoldugunu göstermistir.Anahtar Kelimeler: Yabani emmer bugdayı, gernik bugdayı, T. dicoccoides, T.dicoccon, RAPD-PCR, genetik çesitlik, polimorfizm, UPGMA. It is necessary to characterize genetic diversity of the plant resources for theireffective usage and protection. There are well known various marker systems toanalyse and define the plant genomes. RAPD-PCR is one of the efficiently usedtechnique for distinguishing the genetic variation among the wheat species as wellas the other plants.The objective of this study is molecular characterization of the genetic diversity ofwild emmer wheat and gernik wheat (cultivated emmer) each of which is planted inTurkey by RAPD-PCR technique. For this purpose 11 wild emmer wheats and 8gernik wheats were analyzed. Out of 25 randomly selected primers, 20 were foundto be informative. Of the total 178 amplification products, 85 were polymorphic.Frequency of the polymorphic band patterns were determined as 47.75 % for thewild emmer and gernik wheats. All the data were employed to constructphenograms using an unweighted pair-group method with arithmetical averages(UPGMA). The UPGMA analysis indicated as a result that the lowest similarity wasbetween gernik wheat recorded as TUR 02456 and wild emmer wheat recorded asTUR 03399, whereas, the genetic distance between two gernik wheats which arerecorded as TUR 03562 and TUR 03564 was the highest.Keywords: Wild emmer wheat, gernik wheat, T. dicoccoides, T. dicoccon, RAPDPCR,genetic diversity, polymorphism, UPGMA.
