Abstract
Bu çalışmada, Türkiye'nin farklı bölgelerinde bulunan sıcak su kaplıcalarından alınan su ve çamur örneklerinden termofilik özellik gösteren toplam 130 izolat elde edildi. Konvensiyonel ve moleküler yöntemler kullanılarak yapılan analizler sonucunda; Bacillus, Anoxybacillus, Aeribacillus, Enterococcus, Exigobacterium ve Paenibacillus cinslerine ait toplam 11 farklı tür tespit edildi. Elde edilen veriler incelendiğinde; test suşlarının, genellikle %2-4 tuz konsantrasyonunda, pH 7-9'da ve 45-55°C sıcaklık değerlerinde optimum üreme potansiyeline sahip olduğu belirlendi. Ayrıca termofilik bakterilerin tamamının Gram, Endospor (EA 9 ve EA 10 hariç), katalaz ve oksidaz pozitif (EA 9 hariç), hareketli, çubuk (EA 9 hariç) formda olduğu tespit edildi. 16S rRNA sekans analizi sonucunda tüm izolatların %99 benzerlik yüzdesine sahip olduğu gözlemlendi. Tanılanan 11 farklı türün endüstriyel enzim üretme potansiyelleri petri plak yöntemi kullanılarak incelendi. Elde edilen veriler analiz edildiğinde: EA 4 ve EA 8'in güçlü proteaz, EA 2 ve EA 4'ün güçlü amilaz; EA 2-5, EA 7 ve EA 8 kodlu izolatların güçlü lipaz, EA 2, EA3, EA 7 ve EA 8'in ksilanaz ve EA 6 kodlu izolat ise selülaz enzimi üretme potansiyeli bakımından pozitif olduğu belirlendi. Tüm bu veriler göz önüne alındığında, test suşlarının endüstriyel enzim üretme potansiyellerinin oldukça yüksek olduğu, bu izolatlardan elde edilecek enzimlerin biyoteknolojik proseslerde yaygın kullanım alanı bulacağı düşünülmektedir. In this study, a total of 130 isolates showing thermophilic characteristic from water and sludge sapmles attained from thermal springs found in different regions in Turkey was obtained. As a result of analyses by using conventional and molecular methods, a total of 11 different species which would belong to Bacillus, Anoxybacillus, Aeribacillus, Enterococcus, Exigobacterium and Paenibacillus was determined. When the obtained data were examined, it was determined that the test strains have usually optimum reproductive potency at 2-4% salt concentration, pH 7-9 and tempetature of 45-55 °C. It was also detected that all of the thermophilic bacteria were in the form of grams, endospores (except EA 9 and EA 10), catalase and oxidase positive (except EA 9), mobile, rod (except EA 9). As a consequence of 16S rRNA sequence analysis, it was observed that all isolates had 99% similarity. Afterwards, industrial enzyme-producing potentials of 11 different species identified were examined by using petri plate method. When the obtained data were analyzed, the coded isolates of EA 4 and EA 8 for strong protease-producing potential, EA 2 and EA 4 for strong amylase-producing potential, EA 2-5, EA 7 and EA 8 for strong lipase-producing potential, EA 2, EA 3, EA 7 and EA 8 for xylanase-producing potential and EA 6 for cellulase-producing potential were designated to be positive. When all these data are taken into consideration, it is considered that the test strains have a high industrial enzyme-producing potential and the enzymes obtained from these isolates will be widely used in biotechnological processes.
