Abstract
Bu çalışmada, ilaç etken maddesi olarak sentezlenmiş benzoik asit, benzimidazol ve ß-metil-ß-nitrostiren türevleri olmak üzere üç farklı gruptan nitrolu bileşiklerin genotoksik potansiyelleri umu-mikroplak test sistemi ile değerlendirilmiştir. SOS indükleme aktivitelerinin değerlendirildiği umu-mikroplak test sisteminde, nitrolu bileşiklere hassas olarak geliştirilmiş, nitroredüktaz (NR) enzimini normalden fazla ifade eden Salmonella typhimurium NM1011, O-asetiltransferaz (O-AT) enzimini normalden fazla ifade eden S. typhimurium NM2009 ve nitroredüktaz (NR) enzimiyle O-asetiltransferaz (O-AT) enzimini birlikte normalden fazla ifade eden S. typhimurium NM3009 suşları kullanılmıştır. Enzim assay ortamında klorofenol-red-ß-D-galaktopiranosid (CPRG) substrat olarak ve genotoksik ajan olduğu bilinen 4-nitroquinoline 1-oxide (4NQO) de pozitif kontrol olarak kullanılmıştır.Tüm bileşikler için, üç ayrı suşta, umuC gen ifadesinin indüksiyonu hemen hemen aynı bulunmuştur. Test edilen bileşikler, nitrolu bileşiklerin metabolik aktivasyon mekanizmasında görevli nitroredüktaz ve O-asetiltransferaz enzimlerini normalden fazla ifade eden bu üç suşta belirgin bir indüksiyona neden olmamıştır. Bileşiklerin hiçbirinin genotoksik aktivitesinin olmaması ilaç adayı olarak değerlendirilmelerinde çok önemli bir veridir.Anahtar Kelimeler: Nitrolu bileşik, ilaç, umu-mikroplak testi, genotoksisite. In this study, genotoxic potentials of some nitro compounds (some benzoic acid, benzimidazole and ß-methyl-ß-nitrostyren derivatives) which are designed as drug evaluated with umu-microplate test system. Evaluation of the SOS inducing activity of the tested compounds was examined with the umu-microplate test system using Salmonella typhimurium NM1011 (overexpressed nitroreductase), S.typhimurium NM2009 (overexpressed O-acethyltransferase) and S.typhimurium NM3009 (overexpressed O-acethyltransferase and nitroreductase) strains which are sensitive to nitro compounds. Chlorophenol red- ? -D-galactopyranoside (CPRG) was used as substrate in the enzyme assays and also the well-known genotoxic nitro compound, 4-nitroquinoline 1-oxide (4NQO), was the positive control in the test.For all compounds and for all strains, the induction of umuC gene expression was found to be almost the same. The derivatives tested didn?t caused an evident induction in all strains overexpressed nitroreductase and O-acethyltransferase enzymes which have a role in metabolic activation mechanism of nitro compunds. None of the derivatives show genotoxic effects, a very important data, making them a potential drug candidate.Keywords: Nitro compounds, drug, umu-microplate test, genotoxicity.
