Abstract
Redoks proteinler ile elektrotlar arasındaki doğrudan elektron transferinin incelenmesi duyarlı biyosensörlerin geliştirilmesi için oldukça önemlidir. Fakat, redoks proteinlerin çoğu klasik elektrotlar üzerinde doğrudan elektrokimyasal davranış sergileyemezler. Çünkü redoks aktif ?hem? grupları protein içinde derine gömülüdür. Redoks proteinlerin doğrudan elektron transferini kolaylaştırmak için, elektrot yüzeyleri proteinlerin adsorpsiyonuna uygun en iyi yönlenmeyi sağlayacak şekilde modifiye edilmelidir.Kendiliğinden düzenlenen tek tabakalar (SAM) sağlam ve kontrol edilebilir ara yüzeyler oluşturmak için kullanılan bir yöntemdir. Ayrıca polianilin (PANI) gibi iletken polimerler iyi iletkenlikleri, biyouyumluluklerı ve kolay hazırlanabilir olmaları nedeniyle biyoanalitik sensörlerin dizaynı için kullanılırlar.Bu çalışmada, kendiliğinden düzenlenen tek tabaka/polianilin (SAM/PANI) kaplı altın elektrot üzerine tutuklanmış hemoglobinin (Hb) elektrokimyasal davranışı incelenmiştir. Modifiye edilmiş elektrodun hidrojen peroksit tayini için uygulaması gerçekleştirilmiştir. Kendiliğinden düzenlenen tek tabaka oluşturmak için alkil zincir uzunluğu farklı iki SAM tipi kullanılmıştır (11-merkaptoundekanoik asit, MUA ve 3-merkaptopropionik asid, MPA). Uygun alkantiyol seçiminden sonra, polimerizasyon yöntemi, polimerik film kalınlığı, kendiliğinden düzenlenme süresi, Hb derişimi, pH etkisi ve tarama hızı etkisi gibi optimum çalışma koşulları incelenmiştir. Elektrokimyasal empedans spektroskopisi (EIS), taramalı elektron mikroskopisi (SEM) ve atomik kuvvet mikroskopisi (AFM) ile modifiye edilmiş elektrotlar karakterize edilmiştir.Anahtar Kelimeler: Hemoglobin, SAM, polianilin, biyosensör, hidrojen peroksit. The investigation of the direct electron transfer between redox proteins and electrodes are of great importance for the development of sensitive biosensors. Unfortunately, most of the redox proteins cannot exhibit direct electrochemical response at conventional electrodes because redox active heme groups are deeply buried inside the protein. In order to facilitate direct electron transfer of redox proteins, it is important to modify electrodes for adsorbing proteins in optimal orientation.Self assembled monolayer (SAM) is a common technique to provide relatively rigid and controllable interfaces. Also conductive polymers such as polyaniline (PANI) are often chosen for the design of bioanalytical sensors because of their good conductivity, biocompability and ease of preparation.In this work, the electrochemical behaviour of hemoglobin (Hb) immobilized on self-assembled monolayer/polyaniline (SAM/PANI) coated gold electrode was investigated. The application of the modified electrode was performed for hydrogen peroxide (H2O2) sensing. Two different SAMs with different alkyl chain length were used (11-mercaptoundecanoic acid, MUA and 3-mercaptopropionic acid, MPA) to form self-assembled monolayer. After choosing the appropriate alkanethiol, optimum working conditions such as; the polymerization method, polymeric film thickness, self-assembling time, concentration of Hb, immobilization time of Hb, the effect of pH and the effect of scan rate were investigated. Electrochemical impedance spectroscopy (EIS), scanning electron microscopy (SEM) and atomic force microscopy (AFM) were used to characterize the modified electrodes.Keywords: Hemoglobin, SAM, polyaniline, biosensor, hydrogen peroxide
