Abstract
Tez kapsamında reaktif formda, hidrofobik, monodispers ve gözenekli poli(4-klorometilstiren-co-divinilbenzen) partiküller ?çok basamaklı mikrosüspansiyon polimerizasyonu? ile sentezlenmiş ve boronat afinite kromatografisinde kesikli sistemde destek materyali olarak kullanılmıştır.Reaktif klorometil grubuna sahip, 6 µm boyutundaki monodispers ve gözenekli poli(CMS-co-DVB) partiküllere bazik ortamda 3-aminofenilboronik asit, APBA, nükleofilik sübstitüsyon reaksiyonu ile bağlanmıştır. Çalışmada reaktif klorometil grubuna sahip partiküller ile çalışılmasının nedeni, polimerik yapıya seçilen ligandın kolaylıkla bağlanabilmesidir. Poli(CMS-co-DVB) partiküllerin boyut özellikleri ve yüzey morfolojisi, taramalı elektron mikroskobu, gözeneklilik özellikleri HPLC sisteminde ters-büyüklükçe ayırma kromatografisi, yüzey alanı, BET sistemi ile tayin edilmiştir. APBA bağlı poli(CMS-co-DVB) partiküllerin ligand içeriği 75 mg APBA/g polimer olarak bulunmuştur.Sentezlenen APBA bağlı poli(CMS-co-DVB) partiküller boronat afinite kromatografisi yoluyla kesikli sistemde diol gruplu biyomoleküllerin izolasyonu için kullanılmıştır. Model molekül olarak askorbik asit (AA) ve ribonükleik asit (RNA) seçilmiştir. AA ve RNA için maksimum denge adsorpsiyon kapasiteleri sırasıyla 147 mg AA/g polimer ve 134 mg RNA/g polimer olarak bulunmuştur. Bu değerler diol boronik asit etkileşiminin maksimum olduğu pH 8.5 değerinde elde edilmiştir. RNA ve AA için maksimum desorpsiyon oranları sırasıyla % 65 ve % 75 olarak bulunmuştur. Bu değerler deriştirilen sorbentin, boronat afinite kromatografisi yöntemiyle biyomolekül izolasyonunda başarıyla kullanılabileceğini göstermiştir.Anahtar Kelimeler: Monodispers-gözenekli partiküller, CMS, APBA, adsorpsiyon, boronat afinite kromatografisi, RNA, Askorbik asit. In this study, hydrophobic, monodisperse and macroporous poly(4-chloromethylstyrene-co-divinylbenzene) particles in the reactive form were synthesized by multistage-microsuspension polymerization and used in batch fashion boronate affinity chromatography as a support material. The boronic acid functionalized ligand, 3-aminophenylboronic acid (APBA), was covalently attached onto the monodispers and macroporous poly(CMS-co-DVB) particles in size 6 µm that have reactive chloromethyl group, in basic medium via the direct nucleophilic substitution reaction. In the study because of working with particles have chloromethyl group, selected ligand can easily be attached onto polymeric structure. Size properties and surface morphology, porosity characteristics and surface area of poly(CMS-co-DVB) particles were performed by scanning electron microscopy, inverse-size exclusion chromatography and BET system, respectively. Ligand content of APBA attached poly(CMS-co-DVB) beads was found as 75 mg APBA/g polymer.APBA attached poly(CMS-co-DVB) beads were used as stationary medium for isolation of diol carrying biomolecules in batch fashion by means of boronate affinity chromatography. Ascorbic acid (AA) and ribonucleic acid (RNA) were selected as model molecules. Maximum equilibrium adsorption capacities of RNA and AA were found as 134 mg RNA/g polymer and 147 mg AA/g polymer, respectively. The maximum desorption ratios of RNA and AA were found as % 65 and % 75, respectively. These values show that synthesized sorbents can successfully be used for isolation of biomolecules by boronate affinity chromatography method.Keywords: Monodisperse-macroporous particles, CMS, APBA, boronate affinity chromatography, RNA, Ascorbic acid.
