Abstract
Bu çalışmada doğal kaynaklardan izole edilen yeni bir mikrobiyal kaynaktan fitaz üretimi amaçlanmıştır. Mikroorganizma izolasyonu için ahır ve kümes çevrelerinden alınan 12 farklı toprak örneği kullanılmıştır. İzolatlar arasından K12 kodlu izolatın en yüksek fitaz üreticisi olduğu saptanmıştır. 16S rRNA dizi analizi sonucunda K12 izolatının Bacillus subtilis türüne ait olduğu ortaya çıkmıştır. Yüksek enzim üretimini sağlamak amacıyla K12 izolatının çeşitli üretim parametreleri optimize edilmiştir. Üretim ortamında en uygun karbon kaynağının glukoz ve en uygun azot kaynağının ise amonyum nitrat (NH4NO3) olduğu tespit edilmiştir. Glukoz, amonyum nitrat ve sodyum fitat varlığında 150 rpm çalkalama hızında, 72 saat 300C'de inkübasyonun fitaz üretimi için en uygun koşul olduğu belirtilmiştir. Optimize edilen üretim koşullarında elde edilen fitaz enzimi pH 6.5'da 600C'de 1:9 enzim substrat oranında en yüksek aktiviteye sahiptir. MgCl2, CuCl2, ZnCl2 ve CoCl2 çözeltileri ve EDTA fitaz aktivitesinde kayba neden olurken CaCl2 çözeltisinin aktiviteyi artırıcı yönde etkisi olduğu tespit edilmiştir. Etanol ile kısmi olarak saflaştırılan enzimin SDS-PAGE jel elektroforezi sonucunda yaklaşık olarak 25 ve 17 kDa'luk iki farklı bant elde edilmiştir. In this study, the production of phytase is aimed from a new microbial source that was isolated from natural sources. 12 different soil samples that were taken from barn and coop surroundings are used for the isolation of microorganism. K12 coded isolate among whole isolates is determined to be the highest producer of phytase.As a result of 16S rRNA sequencing analysis,K12 isolate was found Bacillus subtilis. The various production parameters of K12 isolate were optimized in order to ensure high enzyme production. In production medium, glucose was founded as the most suitable carbon source and ammonium nitrate (NH4NO3) was found to be the most suitable nitrogen source. In the presence of glucose, ammonium nitrate and sodium phytate and at the agitation speed of 150 rpm, 72 hours incubation at 30oC is said to be the most suitable conditions for the production of phytase. Phytase enzyme that was obtained from optimized production conditions, at pH 6.5 at 60oC and enzyme substrate ratio of 1:9 has the highest activity. MgCl2, CuCl2, CoCl2, ZnCl2 solutions and EDTA causing a loss of phytase activity whereas CaCl2 solution has been found to be effective as an activity enhancer.As a result of SDS-PAGE gel electrophoresis of enzyme that was partially purified with ethanol, approximately 25 and 17 kDa weight two different bands were obtained.
