Abstract
Bu çalışmada, Ankara Üniversitesi Mühendislik Fakültesi Gıda Mühendisliği Bölümü Kültür Koleksiyonu'nda bulunan 154 adet laktik asit bakteri suşundan sünme özelliklerine bağlı olarak seçilen 10 adet suşun Ekzopolisakkarit (EPS) üretimleri incelenmiştir. EPS üretimini etkileyen fermantasyon koşullarından pH (5.0, 6.0, 7.0), sıcaklık (20°, 30°, 37°C), inkübasyon süresi (48, 120, 192 saat) ve farklı içeriğe sahip besiyerlerin de EPS üretim miktarlarına etkisi incelenmiştir.Çalışma kapsamında incelenen laktik asit bakterisi suşlarından 4 tanesinin (E9, E10, E2, ve E7) yüksek EPS üretim yeteneklerine (sırasıyla, 515.48 mg/L, 512.81 mg/L, 445.65 mg/L ve 444.88 mg/L) sahip oldukları belirlenmiştir. EPS üretimi için en uygun pH 6.0, en uygun sıcaklık 30°C olarak belirlenmiş; en yüksek EPS üretimi için inkübasyon süresi suşlara göre değişiklik göstermiştir. Besi ortamına glikoz ilavesi tüm suşlarda, laktoz ilavesi E1, E6, E7, E8, E9, E10 suşlarında EPS üretimini teşvik etmiş; mineral katkıların önemli bir etkisi belirlenememiştir. In this study, 10 lactic acid bacteria strains?s EPS production examined which are selected from 154 bacteria in Food Engineering Department of Ankara University collection about their ropiness properties. The fermantation conditions from that effect EPS production pH (5.0, 6.0, 7.0), temperature (20°, 30°, 37°C), incubation time (48, 120, 192 hours) and EPS production quantities of medium with different contents were investigated.Lactic asit bacteria strains were examined within the scope of study, it was determined that in four of them (E9, E10, E2, and E7) have high EPS production capabilities (respectively, 515.48 mg/L, 512.81 mg/L, 445.65 mg/L ve 444.88 mg/L). For EPS production, it was determined that the optimum pH was 6.0, optimum temperature was 30°C. For the highest EPS production, incubation time varied according to strains. In medium the addition of glucose in all strains, the addition of lactose in E1, E6, E7, E8, E9, E10 strains encourage the EPS production; mineral additives were not identified as an important effect.
