Abstract
Aktif maddesi ibrutinib olan Imbruvica®, MCL (mantle cell lymphoma), CLL (chronic lymphocytic leukemia) ve WM (Waldenström's macroglobulinemia) kanser türleri için geliştirilmiş ve FDA tarafından onaylanmıştır. İbrutinib ülkemize ithalatla tedarik edilmektedir. Bu tezin amacı İbrutinib için mevcut patentleri ihlal etmeyen yeni bir sentez yönteminin geliştirilmesi ve makro ölçeğe uygulanabilirlik açısından metodun optimizasyonudur. Bu amaçla ilk olarak 4-fenoksibenzoik asit ve malononitrilin kondenzasyonu ile 2-(hidroksi(4-fenoksifenil))metilen malononitril elde edildi ve takiben elde edilen ürün MsCl ile muamele edilerek ilgili mesilat esterine dönüştürüldü. Mesilat esterinin NH2NH2.xH2O ile muamelesi ile 5-amino-3-(4-fenoksifenil)-1H-pirazol-4-karbonitril elde edildi ve bu bileşiğin de formamit ile kondenzasyonu ile 3-(4-fenoksifenil)-1H-pirazolo[3,4-d]pirimidin-4-amin sentezlendi. Pirazolopirimidin'in (S)-1-Boc-piperidin-3-ol metansülfonat ile sübstitüsyonu ile (R)-1-(1-Boc-piperidin-3-il)-3-(4-fenoksifenil)-1H-pirazolo[3,4-d]pirimidin-4-amin elde edildi. Bu üründen Boc grubunun HCl/dioksan ile uzaklaştırılması ile elde edilen amin hidroklorür, DİPEA ve akriloil klorür ile reaksiyona sokularak hedef bileşik olan İbrutinib'e ulaşıldı. Ayrıca, heterojen katalizör olarak G-Ni/Pd kullanılarak gerçekleştirilen Suzuki kenetleme reaksiyonlarıyla ibrutinib sentezlerinin iki ara kademe ürünü için kısa ve yüksek verimli sentez yöntemleri geliştirildi. Imbruvica®, containing ibrutinib as an active substance, was developed for the cancer types MCL (mantle cell lymphoma), CLL (chronic lymphocytic leukemia) and WM (Waldenström's macroglobulinemia) and approved by the FDA. Ibrutinib is supplied with imports in our country. The aim of this PhD thesis is to develop a new synthetic method which does not violate existing patents for Ibrutinib and to optimize the method in terms of applicability to the macro scale. For this purpose, firstly 2- (hydroxy(4-phenoxyphenyl))methylene malononitrile was obtained from the condensation of 4-phenoxybenzoic acid and malononitrile and then was converted to the corresponding mesylate ester by treatment with MsCl. Treatment of the mesylate ester with NH2NH2.xH2O gave 3-(4-phenoxyphenyl)-1H-pyrazolo-4-carbonitrile which was converted to 3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidine-4-amine by condensation with formamide. Substitution of the pyrazolopirimidine with (S)-1-Boc-piperidin-3-ol methanesulfonate afforded (R)-1-(1-Boc-piperidin-3-yl)-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidine-4-amine. The amine hydrochloride obtained by removal of the Boc group with HCl/ dioxane was reacted with DIPEA and acryloyl chloride to give the target compound, Ibrutinib. Furthermore, a short and efficient preparation method was performed for two key intermediate products of ibrutinib synthesis by Suzuki coupling reactions using G-Ni / Pd as heterogeneous catalysts.
