Abstract
Gelmesi muhtemel bir uyarıcıyı önceden öngörerek gerekli tepki ya da tepki kurulumlarını ortaya koymak, organizma için son derece yaşamsal olan bir davranıştır. Bir uyarıcının öngörülmesi sonucu geliştirilen beklenti, hem algılama süreçleri sırasında hem de bu uyarıcıya karşı verilecek motor tepkilerin seçilmesi aşamalarında önemlidir. Bu çalışmanın amacı, insan gibi bilişsel kapasitesi üst düzey olan canlıda, beklentinin elektrofizyolojisini incelemektir. Çalışmada beyin elektrofizyolojisi, olay-ilişkili potansiyel (OİP) ve olay-ilişkili osilasyonlar (OİO) temelinde incelenmiştir. Katılımcılar sağlıklı ve gönüllü erken yetişkin dönemde, üniversite eğitimi gören 33 bireyden (18 kız ve 15 erkek) oluşmuştur. Beklenti tüm paradigmalarda 4. uyarıcıdan sonra gelen 5. uyarıcının atlanması suretiyle oluşturulmuştur. Beklenti için dört farklı paradigma hazırlanmıştır. Bu paradigmaların ilk ikisi saf ses uyarıcılarının (S1/BP, S4/BP), son ikisi ise semantik uyarıcıların (M1/BP ve M4/BP) kullanılmasını içermiştir. Saf sesler 0.5 sn uzunluğunda ve 65 dB ses basınç düzeyindeki sinüs dalgalarından oluşmuştur (1500 Hz, 2000 Hz, 2500 Hz ve 3000 Hz). Semantik uyarıcılar ise bir şiirin mısraları olmuştur. Şiirin kullanıldığı metin paradigmalarında uyarıcıların uzunlukları ve ses basınç düzeyleri, bürünsel özellikler doğrultusunda, değişken olmuştur (62-65 dB). Bu çalışmada katılımcının görevi, atlanan 5. uyarıcının gelme anını tahmin etmek olmuştur. Elektrofizyolojik kayıtlar, sinterli gümüş klorürlü elektrotlar üzerinden 32 kanalı Synamps preamplifikatörü kullanılarak alınmıştır (referans: birleştirilmiş mastoid elektrotları). Alınan kayıtlar çevrim-içi olarak 0.1 - 100 Hz (3 dB down, 12 dB / oktav) frekans aralığında filtrelenmiştir (A/D oranı 500 Hz). Diskriminant analizleri paradigma grubunu yordamada Fz ve Cz elektrot lokasyonlarında N100 bileşeninin; Pz elektrot lokasyonunda ise P300 ve N200 bileşenlerinin genliklerinin etkili olduğunu göstermektedir. Bu bulgular N100 ve P300 ile ilgili literatür bulgularını desteklemektedir. N100'e ilişkin bulgular, bu bileşenin pasif dikkatin yanı sıra dolaylı şekilde aktif dikkat ile de ilgili olabileceğini ortaya koymuştur. P300 bileşeniyle ilgili olarak, 2. uyarıcıdan 4. uyarıcıya doğru Pz elektrot lokasyonundan elde edilen P300 bileşeninin genliği artmıştır. Bu bulguları, atlanan uyarıcı yaklaştıkça katılımcıların dikkatlerini daha fazla yoğunlaştırdıkları ve hazır oluş içine girdikleri yolunda yorumlamak mümkündür. N400 bileşeninin genliği ise frontosentral bir yayılım göstermekte ve atlanan uyarıcı yaklaştıkça azalmaktadır. Bu bulgular, N400'ün bellek içi arama-tarama işlemlerini temsil ettiğine işaret etmektedir. Atlanan uyarıcı için alfa ve teta bandlarında faz düzenliliği (phase ordering) gözlenmektedir. Bu bulgu, katılımcıların, atlanan uyarıcının gelme anında bu uyarıcıyı, içsel olarak üretebildiklerini göstermektedir. Toplam süre göz önünde tutularak yapılan güç spektrumu analizlerinde 2. uyarıcıdan 4. uyarıcıya doğru teta tepkisi gücünde bir azalma görülürken, alfa 1 tepkisi gücünde bir artış görülmüştür. Sadece uyarıcı öncesi dönem göz önünde tutularak oluşturulan güç spektrumlarında ise hem teta hem de alfa 1 tepkisi güçlerinde bir artış olmaktadır. Toplam süre ve uyarıcı öncesi dönemle ilgili bu sonuçlar, atlanan uyarıcıya doğru beynin bir hazır oluşluk ve beklenti içine girmesinin göstergesi olarak yorumlanabilir. Anticipating incoming events and exhibiting response or response sets are crucial to the survival of organisms. The expectancy that is developed when anticipating an event is important both for processes related to the perception of the event and processes related to the selection of the appropriate motor response. The aim of the study was to investigate the electrophysiological basis of expectation in an organism with a high level of cognitive capacity, such as in the humans. Brain's electrophysiology was studied in the present study in the form of event-related potentials (ERPs) and event-related oscillations (EROs).The sample consisted of 33 healthy, right handed university students (18 females and 15 males). To create expectancy effect, the 5th stimulus after every 4th stimulus was omitted in all the following four paradigms. In two of the paradigms (S1/BP, S4/BP), sinusoidal pure tones were used as stimulus. In the other two paradigms (M1/BP and M4/BP), meaningful stimuli were used. Pure tones had 0.5 s duration and 65 dB sound pressure level. In S1/BP, pure tones were in one of the following frequencies: 1500 Hz, 2000 Hz, 2500 Hz ve 3000 Hz. In S4/BP, all four frequencies were used in four different patterns. For the meaningful stimuli, four verses of a poem were used. In M1/BP one verse of the poem was used; in M4/BP, all four verses were used. Duration of the words and the sound pressure levels could change for each word (62-65 dB) because of prosody effect. The task of the participants was to guess the time point of the 5th omitted stimulus. Electrophysiological recordings were taken using a 32-channel preamplificator. The electrodes were made of sintered Ag-Ag/Cl and referenced to the linked mastoids. Recordings were filtered online between 0.1 Hz and 100 Hz (3 dB down, 12 dB / oct) at an analog-digital conversion rate of 500 Hz.Discriminant analyses showed that N100 had the highest predictive value for levels of the paradigm at Fz and Cz electrode locations; and P300 and N200 components had the highest predictive value at the Pz electrode location. These findings support the findings in the related literature according to which the N100 component is related to passive and indirectly to also the active attention. P300 amplitude increased from the 2nd stimulus to the 4th stimulus at the Pz electrode location. This change suggested an increased focused attention and readiness to respond as the time of the omitted stimulus approached. N400 component showed a frontocentral distribution and its amplitude decreased as the 4th stimulus approached. These findings suggested that N400 reflects complex stimulus evaluation and processes related to memory search and scan. Phase-reordering was found at the alpha and theta bands. This indicates that participants can produce omitted stimulus internally at the time point where stimulus should have occurred. Power spectral analysis of the total recording time showed that theta power decreased from 2nd to the 4th stimulus. However, alpha 1 power increased from the 2nd to the 4th stimulus. However, analyses covering only the prestimulus period showed that both theta and the alpha power increased from the 2nd stimulus to the 4th stimulus. These results show that increased readiness to respond and expectation state of the brain as the time point of the omitted stimulus approaches can be represented not only by the by the event-related potentials but also the event-related oscillations of the brain.
