Türkiye`nin göller bölgesinden toplanan yonca (Medicago sativa L.) populasyonlarının karyotip karakterizasyonu

Abstract
Yonca (Medicago sativa L.), dünyada ve Türkiye'de en önemli baklagil yem bitkisidir. Özellikle Türkiye'de çok fazla ekonomik öneme sahiptir. Yonca, dünyada 60 türe sahiptir ve bu türlerin 30 tanesi Türkiye'nin farklı bölgelerinde yayılmıştır. Türkiye'nin Göller Bölgesi'nde benzer iklim koşullarında bulunan yonca populasyonları bir gen kaynağı olarak geliştirilmemiş, bu nedenle o bölgeyi temsil eden 25 populasyon, üzerinde sitogenetik çalışmalar yapılmak üzere 22 farklı bölgeden toplanmıştır. Serada saksılarda yetiştirilen tek bitkilerden klonla çoğaltımla elde edilen bitkilerden alınan kök ucu meristemlerine ?-bromonaftalin doymuş çözeltisinde ve 0.002 M 8-hydroxiquinolin ön işlem yapılmış ve sonra kök uçları aceto-iron-hematoxylin ile boyanmıştır. Ölçülen kromozomal parametreler; Kısa Kol, Uzun Kol, Toplam Kromozom Boyu, Kol Oranı, Sentromer Indeksi (CI), Kromozomların Nispi Uzunluğu (RL%) ve Karyotipin toplam formu yüzdesi (TF%)dir. Karyotip analizi, tüm populasyonların tetraploid (2n= 4x= 32) olduğunu göstermiştir. Kromozomların boyları küçüktür. Kromozom boyları 1.69-6.20 mikron arasında değişmektedir. B- kromozomları ve heteromorfik kromozomlar belirlenmiştir.Bu bölgelerde yoncanın heterojen populasyonları olduğu kanıtlanmıştır. Araştırmadan elde edilen veriler, tamamen faktöriyel tesadüf desene (CRD) dayalı 5 hücre replikasyonlu olarak analiz edilmiştir. ANOVA, değerlendirilen tüm özellikler için, populasyonlar arasındaki göze çarpan farklılıkları göstermiştir. Karyotip formülüne dayalı olarak populasyonlar 8 farklı sınıfta kategorize edilen 2 tip kromozoma(m, sm) sahiptir. Farklı parametreler, örneğin DRL (Nispi kromozom boyunun aralık farkı ), DI (Dağılım İndeksi), %TF (Karyotipin toplam formu yüzdesi), %CV (Varyasyon Katsayısı), Stebbins ve Romero-Zarco kategorileri kullanılarak karyotip simetri değerlendirilmesi yapılmıştır. Korelasyon katsayısı sonuçları, uzun kolun en fazla toplam kromozom boyu ile ilişkili olduğunu ve kol oranının ise en düşük kısa kol ile ilişkili olduğunu göstermiştir. Kluster analizi UPGMA metoduna dayalı olarak yapılmış, eucldean kare mesafe metodu ile belirlenmiş olup: dendogram 4 grup göstermiştir. Temel bileşenlere analizi,ilk üç bileşenin, verilerin variyasyonunun %92.67'sini kapsadığını göstermiştir. İlk 2 PC'ye göre de populasyonlar 4 grupta sınıflandırılmıştır. Hüyük-2 populasyonu birey grubu oluşturmuş ve populasyonların geri kalanı 3 grupta kümelenmiştir. Alfalfa (Medicago sativa L.) is the one of the important and economic forage legume crops in the world and Turkey. Alfalfa has 60 species which 30 annual and perennial species were distributed in different regions of Turkey. The alfalfa populations of Lake Regions of Turkey as gene resources have not been improved; therefore, 25 representatives alfalfa populations were collected from 22 different locations of this province with similar climatic conditions for cytogenetical studies. Root tips meristems obtained from plants of similar age that originated by vegetative reproduction from a single parent from plants grown under greenhouse conditions, were pretreated with saturated solution of ?-bromonaphthalene and 0.002 M 8-Hydroxiquinolin before staining with aceto-iron-hematoxylin. The measured chromosomal parameters were; Short arm, Long arm, Total length of chromosomes, Arm Ratio, Centromere Index (CI), Relative Length (RL%) and Total Form Percentage (TF%). Karyotype analysis showed that all populations were tetraploid (2n=4x= 32). The chromosomes are small, ranging from 1.69-6.20 microns in length. B chromosome and heteromorphic chromosomes were established. Due to these phenomenons, the existence of heterogeneous in alfalfa populations were demonstrated from these locations. The resultant data were analyzed based on completely randomized design (CRD) factorial with 5 replicates of cells. ANOVA indicated marked differences among Populations for most of the evaluated traits. On the basis of karyotypic formula, the populations had 2 types of chromosomes (m, sm), categorizing them in 8 different classes. Assessment of karyotype symmetry was carried out, using various parameters e.g. DRL, DI, TF%, CV%, Stebbins and Romero-Zarco categories. The result of correlation coefficient showed that the Long arm had the most correlation with Total length of chromosomes, and Short arm had the least correlation with arm ratio. In cluster analysis based on UPGMA method, the distances were determined by eucldean square distance method: dendrogram demonstrated 4 groups. The result of the principal components analysis showed that the first three components covered 92.67% of the data variation. According to the first two PCs, the Populations were categorized into 4 groups as well. the Hüyük-2 population formed individual group and the rest of populations were clustered in 3 groups.
