Abstract
Mevcut çalışmada, 2008 yılında Adana'dan toplanan ve laboratuarda yetiştirilmekte olan pamuk yaprak kurdu Spodoptera littoralis (Boisd.) (Lepidoptera:Noctuidae) popülasyonlarının sentetik piretroitli (gamma-cyhalothrin) ve neonikotenoidli (imidakloprid) insektisitlere karşı gelişen direnç mekanizmalarının belirlenmesi amaçlanmıştır.Gamma-cyhalothrin ve imidaklopride dirençli ırklar elde etmek amacıyla popülasyonlara ilaçlara göre sırasıyla 12 ve 6 nesil boyunca LC75 konsantrasyonunda seleksiyon baskısı uygulanmış ve LC50 bazında aynı ilaçlar için sırasıyla 16.85 ve 9.76 kat hassasiyet azalışı saptanmıştır.Direnç mekanizmasının hangi detoksifikasyon enzim grubu ile ilişkisi olduğunun tespit edilmesi amacıyla S,S,S-tributilfosforotritioat (DEF), piperonyl butoxide (PBO) ve diethyl maleate (DEM) olmak üzere üç farklı enzim inhibitörü sinerjist olarak kullanılmıştır. Sinerjistik çalışmalarda, gamma-cyhalothrin etkisinin incelemesinde, LC50 değerlerine göre DEF, PBO ve DEM için sırasıyla 3.1, 2.1 ve 3.6 kat sinerjizim oranı (SR) tespit edilmiştir. İmidakloprid'le yapılan çalışmaların sonuçları ise sırasıyla 1.7, 1.2 ve 2 kat olarak elde edilmiştir. Biyokimyasal analizler sonucunda, artan direnç oranıyla doğru orantılı olarak, karboksilesteraz, acethylcholinesterase (AChE) ve glutatyon S-transferaz enzim aktivitelerinin anlamlı bir şekilde arttığı tespit edilmiştir. In the current study, it is aimed to determine the resistance mechanisms developed by the Cotton leafworm, Spodoptera littoralis (Boisd.) (Lepidoptera:Noctuidae) populations collected from Adana province in 2008 and being reated under laboratory conditions against synthetic pyrethroids (gamma-cyhalothrin) and neonicotinoids (imidacloprid).A selection pressure was created using LC75 values for 12 and 6 generations for gamma-cyhalothrin and imidacloprid, respectively, in order to obtain insecticide-resistant strains, which revealed sensitivity decrases for 16.85 and 9.76 fold for gamma-cyhalothrin and imidacloprid.In order to detect the relation between the resistance mechanism and detoxification enzyme groups, three enzyme inhibitor, S, S, S-tributilfosforotritioat (DEF), piperonyl butoxide (PBO) and diethyl maleate (DEM) were used as synergists. In the synergistic examination for gamma-cyhalothrin, the effect of above three synergists in LD50 values, synergism ratio (SR) were determind, 3.1, 2.1 and 3.6-fold respectively. The results of studies for imidacloprid were achieved 1.7, 1.2 and 2-fold SR for these synergists, respectively.Biochemical analysis revealed that the activities of carboxylesteras, acethylcholinesteras (AChE) and glutathione-S-transferase were found to increase by the increasing resistance ratio.
