Sıçan karaciğerinde karbon tetraklorür (CCl4) hasarına karşı melatonin ve D vitamininin oksidatif stres ve MMP-9 üzerine in vivo, in vitro ve histokimyasal incelemeler

Abstract
Bu çalışmada CCl4 ile oluşturulan karaciğer hasarına karşı melatonin (MEL) ve D Vitamininin (D Vit) iyileştirici ve koruyucu etkilerinin incelenmesi amaçlanmıştır. Çalışmada, 180-220gr ağırlığında 42 adet Wistar-Albino erkek sıçan kullanıldı. Sıçanlar, kontrol grupları (G1, G2), CCl4 (G3), CCl4+MEL (I) (G4), CCl4+MEL (II) (G5), CCl4+D Vit (I) (G6) ve CCl4+D Vit (II) (G7) olmak üzere 7 gruba ayrıldı. Hepatik fibrozis CCl4 verilerek oluşturuldu. CCl4 enjeksiyonununun başlangıcından itibaren, G4 ve G5'e MEL; G6 ve G7'ye D Vit verildi. CCl4 enjeksiyonunun kesilmesinin ardından, 2 hafta daha G5'e MEL ve G7'ye D Vit uygulamasına devam edildi. Deney sonunda sıçanlardan intrakardiyak kan alınarak, karaciğer dokuları çıkarıldı. Hepatik hasar, kan serum örneklerinin biyokimyasal analizi ve doku örneklerinde histopatolojik incelemelerle değerlendirildi. Doku homojenatlarında glutatyon (GSH), malondialdehit (MDA) ve süperoksit dismutaz (SOD) seviyeleri ölçüldü. MMP-9 ve dezmin aktiviteleri immünohistokimyasal incemelerle saptandı. HepG2 ve Hep3B hücrelerinde canlılık MTT yöntemi ile belirlendi. Sonuç olarak, CCl4 grubunda MDA seviyesinin arttığı GSH seviyesinin azaldığı izlendi. ALT, ALP, GGT, CHOL seviyeleri artarken, BA ve ALB değerlerinin düştüğü gözlendi. Histokimyasal olarak hepatosit dejenerasyonu, lobular dizilim bozukluğu, sinüzoidal dilatasyon, fokal nekrotik alanlar, hiperemi, glikojen kayıpları ve retiküler lif kalınlaşması belirlendi. Portal alan ve sentral ven çevresinde hepatik fibrozis gözlendi. Portal venler arasında köprüleşen fibröz septa izlendi. MMP-9 ve dezmin aktivitelerinde artış saptandı. Melatonin ve D Vitamini uygulamalarının hasarı kısmen telafi edilebildiği görüldü. In vitro olarak ta, hücre canlılığının korunduğu saptandı. Elde edilen bulgulara göre, CCl4 ile oluşturulan karaciğer hasarına karşı hem melatoninin hem de D Vitamininin lipit peroksidasyonunu azalttığı ve antifibrotik rol oynayabildiği ileri sürülebilir. In this study investigation of the curative and protective effects of melatonin (MEL) and Vitamin D (Vit D) against liver injury induced by CCl4 was aimed. 42 Wistar-Albino male rats weighing 180-220g were used in the experiment. Rats were divided into 7 groups: Controls (G1, G2), CCl4 (G3), CCl4+MEL (I) (G4), CCl4+MEL (II) (G5), CCl4+D Vit (I) (G6) and CCl4+D Vit (II) (G7). Hepatic fibrosis was induced by giving CCl4. At the beginning of CCl4 injection, MEL to G4 and G5; Vit D to G6 and G7 were applied. Following the cessation of CCl4 injection, MEL application to G5 and Vit D administration to G7 was continued for two weeks. At the end of the experiment, intracardiac blood was taken and liver tissues were removed. Hepatic damage was evaluated by biochemical analysis of liver serum samples and histopathological examinations in tissue samples. The levels of malondialdehyde (MDA), glutathione (GSH) and superoxide dismutase (SOD) were investigated in tissue homogenates. MMP-9 and desmin activities were performed with immunohistochemical observations. The viability in HepG2 and Hep3B was determined by MTT assay. As a result, increase in MDA level and decrease in GSH content were detected in the CCl4 group. Rise in the levels of ALT, ALP, GGT and CHOL levels, while decrease in the values of BA and ALB were monitored. Hepatocyte degeneration, lobular malalignment, sinusoidal dilatation, focal necrotic areas, hyperemia, glycogen losses and reticular fiber thickening were observed histochemically. Hepatic fibrosis was detected around the portal area and central vein. Bridging fibrous septa was observed between portal veins. Rise in MMP-9 and desmin activities were determined. Relatively compensated damage in liver tissue was observed in melatonin and Vitamin D applied groups. As in vitro, protection in the cell viability were determined. According to the findings obtained, it can be proposed that melatonin as well as vitamin D reduce lipid peroxidation and play antifibrotic role against the liver injury induced by CCl4.
