Abstract
Amaç: Bu çalışmada karadut konsantresinin üretimi ve oda sıcaklığı (20±2 ℃) ile buzdolabı sıcaklığında (4±2 ℃) altmış gün depolanması sırasında depolamanın başlangıcında (0. gün), 20., 40. ve 60. günlerinde bazı fiziksel ve kimyasal özelliklerinde, fenolik profilinde, toplam monomerik antosiyanin içeriğinde, antioksidan ve antimikrobiyal aktivitesinde meydana gelen değişmeleri araştırmak amaçlanmıştır.Yöntem: Kurutulmuş karadut meyvesinde, üretimde ara ürün olan şırada ve konsantrede depolama sırasında bazı fizikokimyasal özelliklerin yanısıra toplam fenolik madde miktarı Folin-Ciocalteu, antioksidan aktivite DPPH• ve ABTS+•, fenolik profili HPLC, antimikrobiyal aktivite kuyu difüzyon yöntemleri ile belirlenmiştir.Bulgular: Depolamanın başlangıcında karadut konsantresinde suda çözünür kuru madde %60,47, kül %4,69, pH 5,15, titrasyon asitliği %0,83, toplam şeker %41,74, HMF 6,10 mg/kg, toplam fenolik madde 550,06 μg GAE/mg, toplam monomerik antosiyanin 107,71 mg/L, IC50 değeri DPPH•'da 10,38 μg/ml ve ABTS+• yönteminde 8,03 μg/ml, L değeri 21,11, a değeri +0,46, b değeri +1,62 olarak belirlenmiştir. Kurutulmuş karadut meyvesinin, şıranın ve karadut konsantresinin araştırmada test edilen mikroorganizmalara karşı antimikrobiyal aktivitesi tespit edilememiştir. Kamferol-3-glikozit karadut konsantresinde, klorojenik asit ise kurutulmuş karadut meyvesinde ve karadut konsantresinde tespit edilmiştir.Sonuç: Karadut konsantresi, toplam fenolik madde ve antioksidan aktivitesi bakımından zengin kaynak olup, fonksiyonel gıda olarak kullanılabilir. Purpose: The objective of this study is to evaluate changes in some physical and chemical properties, phenolic profile, total monomeric anthocyanin amount, antioxidant and antimicrobial activity of black mulberry concentrate during the production and storage at room temperature (20 ± 2 ℃) and refrigerator temperature (4 ± 2 ℃) for sixty days (at 0 th (beginning), 20th, 40th and 60th days).Method: Dried black mulberry, şıra and stored concentrate, were analyzed for some physical and chemical properties. Besides, total phenolic content was estimated using the Folin-Ciocalteu method, antioxidant activity was measured DPPH• and ABTS+• assays, phenolic profile was determined using HPLC, antimicrobial activity was determined well diffusion methods.Findings: At the beginning of storage, in black mulberry concentrate were determined water soluble dry matter 60,47%, ash 4,69%, pH 5,15, titratable acidity 0,83%, total sugar 41,74%, HMF 6,10 mg/kg, total phenolic content 550,06 μg GAE/mg, total monomeric anthocyanin 107,71 mg/L, DPPH• (IC50 value) 10,38 μg/ml, ABTS+• (IC50 value) 8,03 μg/ml, L value 21,11, a value +0,46, b value +1,62. The antimicrobial activity of dried black mulberry fruit, mulberry juice and black mulberry concentrate could not be detected against microorganisms tested in the research. Kaempferol 3-glucoside was detected in black mulberry concentrate, while chlorogenic acid in dried black mulberry fruit and black mulberry concentrate.Results: Black Mulberry concentrate is a good source in terms of total phenolic substance and antioxidant activity and it can be used as a functional food.
