Abstract
Radyasyonun insanlar üzerine zararlı etkileri olmasına rağmen, yüksek seviyedeki iyonize radyasyon varlığında hayatta kalabilen, ekstremofil bakterilerden biri olarak bilinen Deinococcus radiodurans, kuruluk, oksidanlar, Ultra Viole Radyasyon (UVR) gibi aşırı strese olağanüstü karşı koyma yeteneğindedir. Bu çalışma kapsamında, D.radiodurans R1'in direnç fizyolojisinde önemli olduğunu düşündüğümüz hücre içi DrCA enziminin klonlanması, saflaştırılması ve kısmi karakterizasyon çalışmaları yapılmıştır. Ayrıca bakterinin çeşitli dozlarda gama radyasyon uygulaması ve farklı pH değerlerine sahip TGYB besiyerlerinde geliştirilmesi sonucunda radyasyon ve pH ile ilişkili DrCA enzim aktivitelerindeki değişimler belirlenmiştir. Bakterinin en iyi gelişim gösterdiği pH aralığı 6.75-7.75 olarak belirlenmiş ve 24 saat süresince DrCA enziminin en yüksek aktivite değişiminin olduğu pH değeri 7.0 olarak bulunmuştur. Radyasyon uygulaması sonrası DrCA aktivite sonuçlarında, ilk 800 Gy'a kadar aktivitenin 5.4 kat olacak şekilde sürekli arttığı, bu noktadan sonra aktivitede bir düşüş görüldüğü tespit edilmiştir. PCR ile çoğaltılan DrCA geni, pFN2K Flexi vektörüne klonlanmış ve sonuçlar dizi analizi ile de teyit edilmiştir. Sub-klonlama çalışmaları için E. coli DH5α ve protein ekspresyonu için ise E. coli BL21 (DE3) konakları kullanılmıştır Saflaştırma aşamasında MagneGST saflaştırma sistemi kullanılmış ve enzim %94.68 oranında yüksek bir verim ve 494.5 saflaştırma kat sayısı ile saflaştırılabilmiştir. Ayrıca rekombinant E.coli BL21 konağında enzimin spesifik hidrataz aktivitesi 0.109 U/mg iken, saf enzimin spesifik aktivitesi 5 kat artarak 0.539 U/mg olarak belirlenmiştir. Enzimin pH 9.0'de optimum aktivite ve termostabilite gösterdiği sıcaklık 30oC olarak bulunmuştur. Enzimin pH 8.0'de, 25oC'de en yüksek hidrataz aktivitesi gösterdiği saptanmıştır. Saf enzimin esteraz aktivitesi ise eser miktarda (0.0002 U/ml) bulunmuştur. Saf enzim üzerinde metal iyonlarından Zn+2 aktivatör, sülfonamid ise inhibitör etki göstermiştir. Saf enzimin pNP-asetat substrata afinitesinin çok düşük olduğu ve diğer β-DrCA'lar gibi CO2'e yüksek spesifite gösterdiği belirlenmiştir. Although the radiation has harmful effects on humans, Deinococcus radiodurans, known as one of the extremophile bacteria which is capable of surviving at high levels of ionizing radiation, has the ability to resist extreme stresses such as dryness, oxidants, Ultra-Viole Radiation (UVR). In this study, cloning, purification and partial characterization studies of the intracellular DrCA enzyme, which we considered that important in the resistance physiology of D.radiodurans R1, have been conducted. In addition, changes in radiation and pH-related DrCA enzyme activities as a result of gamma irradiation at various doses, and development of the bacterium in TGYB media with different pH values have been identified. The pH range at which the bacteria showed the best improvement was 6.75-7.75 and the pH value at which the highest activity change of DrCA enzyme was found to be 7.0 for 24 hours. DrCA activity data after radiation treatment showed that the activity continuously increased by 5.4 fold, up to the first 800 Gy, which a decrease in activity was observed thereafter. The PCR-amplified DrCA gene was cloned into the pFN2K Flexi vector and the results confirmed by sequence analysis. For gene subcloning studies E. coli DH5α and for protein expression E. coli BL21 (DE3) was used as the host. The MagneGST purification system was used during the purification step and the enzyme was purified with a yield of 94.68% and a purification index of 494.5. Also, the specific hydratase activity of the enzyme in the recombinant E. coli BL21 was 0.109 U/mg, while the specific activity of the pure enzyme was found to be 0.539 U/mg with a 5-fold increase. The temperature at which the enzyme exhibits optimum activity and thermostability at pH 9.0 was found to be 30°C. The esterase activity of the pure enzyme was found in trace amounts (0.0002 U/ml). It has been found that the enzyme exhibited high activity at 25oC and pH 8.0 Among the divalent metal ions, Zn+2 showed activatory and sulfonamide showed inhibitory effect on the pure enzyme. It has been determined that the affinity of the pure enzyme to the pNP-acetate was very low and exhibited high specificity to CO2 as other β-DrCAs.
