Abstract
Kanser, insanoğlu için ölümle sonuçlanan hastalıklardandır. Dünya çapındaki araştırmacılar, sağlıklı hücreleri etkilemeden, seçici olarak kanseri hedef alacak en uygun tedaviyi arıyorlar. Kaliksaren temelli moleküller bu açıdan oldukça umut vericidir ve hedefli tedavi için tercihli sitotoksisiteyi artırmak için modifiye edilebilir. Bu çalışmada, piridinyum grupları ile fonksiyonlandırılmış kalix[4]aren türevlerinin meme kanseri hücreleri üzerindeki tercihli sitotoksitesi incelenmiştir.p-tert-bütilkaliks[4]arenler, farklı konumlarda (2, 3 ve 4 AMP) uygun prosedürlerle piridinyum grubu ile türevlendirildi. İnsan meme adenokarsinoma hücreleri (MCF-7) ve sağlıklı fare fibroblastı hücreleri (L929), maddelerin dozu maximum 500 µM olacak şekilde inkübe edildi. 24 saat inkübasyondan sonra hücre proliferasyonu analizi, XTT metodu ile yapıldı. xCELLigence analiz sistemi ile hücre canlılığı 180 saat boyunca incelendi. Hücre ölüm mekanizmasını incelemek için flow sitometri analizi yapıldı. XTT metodu ile elde edilen IC50 değerleri sırasıyla 2-3 ve 4 AMP için 391.40 µM, 40.96 µM ve 105.52 µM olarak bulundu. xCell sonucuna göre de piridinyum grup 3 pozisyonunda bulunan bileşik, 2 ve 4 pozisyonlarına göre daha sitotoksik bulundu. L929 hücrelerine karşı, hem XTT hem de XCell analizleri sonucunda sitotoksik etkinin bulunmadığı görüldü. Flow sitometri analizi sonucunda hücre ölüm şekli olarak DOX'un %53.2 ile nekroza gittiği 3AMP'nin ise konsantrasyonu 25 µM'dan 50 µM'a yükseldiğinde erken apoptoz popülasyonu %6.5'ten %17.9'a yükseldiği görüldü. Bu çalışmada elde edilen sonuçlar, p-tert-butilkaliks[4]aren'in L929 hücrelerine kıyasla MCF-7'ye karşı tümöre pasif hedefleme yaparak in vitro sitotoksisitesinin, etkinliği kanıtlanmış antikanser ilaçlarına alternatif, etkin ve güvenli kanser tedavisi için umut verici bir yaklaşım sunduğunu ortaya koymuştur. Cancer is one of the diseases that cause death for human beings. Researchers worldwide have been searching for the optimal treatment that will selectively target cancer, without afflicting healthy cells.. The calixarenes based molecules are highly promising in this respect and may be modified to increase preferential cytotoxicity for targeted therapy. In this present study, the effect of derivation of calixarene by pyridinium groups on preferential cytotoxicity in breasr cancer cells were examined.p-tert-butyl calix[4]arenes were functionalized with the pyridinium group by appropriate procedures in different positions (AMP at positions 2,3 and 4). Human breast adenocarcinoma cells (MCF-7) and healthy mouse fibroblast cells (L929) were incubated with samples at maximum concentrations 500 µM. After 24 hours of incubation, the cell proliferation analysis was performed by XTT assay. xCELLigence analysis system were used to observe cell viability for 180 hours. Cell death mechanisms were also observed by flow cytometry.IC50 values were 391.40 µM, 40.96 µM and 105.52 µM for 2-3and 4 AMP, respectively in XTT assay. The compound in the pyridinium group 3 position was also more cytotoxic than the 2 and 4 positions in xCell analysis. Both XTT and xCell analysis revealed no cytotoxic effect against L929 cells. As a result of flow cytometry analysis, it was observed that DOX had necrosis with 53.2% as cell death and when the concentration of 3 AMPs increased from 25 µM to 50 uM, early apoptosis population increased from 6.5% to 17.9%. The results obtained in this study showed that the in vitro cytotoxicity of p-tert-butyl calix[4] aren which performing passive targeting to the tumor against MCF-7 compared to L929 cells offers a promising approach for effective and safe cancer treatment as alternative to proven anticancer drugs.
