Abstract
BEYİNDE ÇİNKONUN SEBEP OLDUĞU HÜCRE ÖLÜMÜNE NİTRİK OKSİT SENTAZ İNHİBİTÖRÜ AMİNOGUANİDİNİN ETKİSİ ÖZET Çinkonun nöronal hiperaktivite ve hücre ölümüne yol açtığına dair çok sayıda çalışma vardır. Bununla birlikte çinko nörotoksisitesine nitrik oksitin (NO) etkisini içeren bir çalışmaya rastlanamamıştır. Bu nedenle sıçanda somatomotor kortekse çinko sülfat verildikten sonra spesifik bir nitrik oksit sentaz (iNOS) enzim inhibitörü olan aminoguanidinin (AG) hipokampusa ait piramidal hücre sayısına olan etkisini tespit etmek üzere sunulan çalışma planlandı. 2 ul hacimdeki çinko sülfat (200 (J.g/kg) sol hemisfere (Bregmanın lateraline) intrakortikal yoldan enjekte edildi. Kontrol grubuna aynı hacimde serum fizyolojik verildi. Tedavi (çinko+ aminoguanidin) grubuna aminoguanidin (100 mg/kg) günde iki kez olmak üzere 7 ve 15 gün süreyle intraperitoneal yoldan verildi. Bu sürelerin sonunda sıçanlar intrakortikal yoldan nötral formalin ile perfüzyona alındı. Hipokampustan elde edilen kesitler Kresyl violet ile boyandı. Stereolojik bir sayım metodu olan optik parçalama yöntemi kullanılarak sol hipokampusa ait toplam piramidal hücre sayısı tespit edildi. Yedi günlük sıçanlarda, sol hipokampusa ait piramidal hücre sayısının kontrol grubunda 186 705 ± 3452.46, çinko grubunda 73 705 ± 1277.46 ve çinko + aminoguanidin grubunda 109 291 ± 1973.44 olduğu bulundu. Gruplar arasındaki fark istatistik açıdan anlamlı idi (p<0.05). Onbeş günlük sıçanlarda, sol hipokampusa ait piramidal hücre sayısının kontrol grubunda 171 960 ± 3348.74, çinko grubunda 71 587 ± 1443.85 ve çinko + aminoguanidin grubunda ise 104 537 ± 2658.31 olduğu tespit edildi. Gruplar arasındaki fark istatistik açıdan anlamlı bulundu (p<0.05). Yedi ve 15 günlük gruplar toplam nöron sayısı yönünden karşılaştırıldığında aradaki fark istatistik açıdan anlamlı değildi (p>0.05). Sunulan çalışma ile sıçanda çinkonun nörotoksik etkisinin spesifik bir NOS inhibitörü olan aminoguanidin tarafından azaltıldığına dair sonuçlar tamamen yeni bulgulardır. Elde edilen bulgular çinkonun merkez sinir sistemindeki nörotoksik etkisinde nitrik oksitin de rol oynadığını ve çinkoya ait bu etkinin NO tarafından artırıldığını göstermektedir. Sıçan IX THE EFFECT OF NITRIC OXIDE SYNTHASE INHIBITOR AMINOGUANIDINE ON THE CELL DEATH INDUCED BY ZINC ADMINISTRATION IN THE BRAIN OF RAT ABSTRACT There are many studies about zinc as a possible cause of neuronal hyperactivity and cell death. However, the present study was designed to investigate the effect of aminoguanidine (AG), which is a specific inhibitor of inducible nitric oxide synthase (iNOS), on Pyramidal cell number after intracortical zinc sulfate administration since not much work has been carried out. The amount of 2ul zinc sulfate (200 ug/kg, i.e.) was applied intracortically on a point lateral to the Bregma on the left hemisphere of rat brain. Control group received the same volume of saline solution. Animals in treatment (zinc+aminoguanidine) group received 100mg/kg AG intraperitoneally twice a day for periods of 7 and 15 days. End of this period, all rats perfused intracardially with neutral formaline. Sections obtained from hippocampus stained with cresyl violet staining. The total number of Pyramidal cell was estimated by using optical fractionator which is a combination of a novel, efficient and unbiased stereological counting method (Optical disector) and an unbiased sampling (fractionator) scheme. The total number of Pyramidal cell was found in the left hippocampus 186 705 ± 3452.46, 171 960 ± 3348.74 in control group, 73 705 ± 1277.46, 71 587 ± 1443.85 in zinc group and 109 291 ± 1973.44, 104 537 ± 2658.31 in treatment (zinc + AG) group in 7 days and 1 5 days rats, respectively. The difference between group was statistically significant (p<0.05). However, there was no significant difference between 7 days and 15 days groups (p>0.05). For the first time, we have found that zinc may have a neurotoxic effect which cause cell death in central nervous system, and specific inhibitor of nitric oxide synthesizing enzyme aminoguanidine may attenuate the cell death induced by zinc administration.
