Ratlarda roziglitazonun mitokondriyal genom üzerindeki etkilerinin moleküler yöntemlerle araştırılması

Abstract
Mitokondrial DNA'da görülen yaygın delesyon 4834 baz uzunluğundadır. Bu delesyon normal yaşlanma sürecinde oksidatif fosforilasyon kapasitesinin azalmasının bir sonucu olarak görülebilir ve aynı zamanda serbest radikallerin üretilmesi ile de ilişkili olabilir. Yaygın delesyon genellikle oksidatif hasarın bir belirteci olarak da kullanılmaktadır. Roziglitazon, Tip II diabet tedavisinde kulanılan tiazolidinedion sınıfına ait bir ilaçtır. Bu sınıf ilaçlar peroksizom proliferatör aktive reseptör-gama'nın ligandı olarak etki gösterirler.Bu çalışmada amaç, farklı rat dokularında yaygın delesyon görülme sıklığının incelenerek roziglitazonun mitokondriyal DNA üzerine olan etkilerinin araştırılmasıdır. Bu amaçla Sprague-Dawley cinsi erkek ratların karaciğer, ince bağırsak, pankreas dokuları ve lenfositlerinden nükleer ve mitokondriyal DNA izolasyonu yapıldı. Kantitatif PCR ile numunelerin nükleer DNA, mitokondriyal DNA ve yaygın delesyon kopya sayıları belirlendi. Yaygın delesyon frekansı ve hücre başına düşen mitokondriyal DNA kopya sayıları hesaplandı.Roziglitazonun tüm dozlarında karaciğer dokusunda yaygın delesyon frekansı kontrol grubuna göre anlamlı olarak düşük bulundu. Bununla beraber ince bağırsak ve pankreas dokularında hücre başına düşen mitokondriyal DNA kopya sayısı roziglitazonun tüm dozlarında kontrol grubuyla karşılaştırıldığında anlamlı olarak daha düşüktü. Aynı zamanda kontrol grupları arasında da yaygın delesyon frekansı ve hücre başına düşen mitokondriyal DNA kopya sayısında istatistiksel olarak anlamlı fark vardı.Sonuç olarak, karaciğer dokusunda yaygın delesyon frekansındaki azalmaya yol açması roziglitazonun oksidan bir etkisinin olmadığını göstermektedir. Fakat pankreas ve ince bağırsaktaki mitokondriyal DNA kopya sayılarınında azalmaya neden olması roziglitazonun oksidan etkisinin olduğunu düşündürebilir.Anahtar Kelimeler: Roziglitazon, Yaygın Delesyon, Oksidatif Stres The common deletion seen at mitochondrial DNA has a length of 4834 base pair. This deletion could be seen as a result of the reduction of oxidative phosphorylation capacity in the normal aging process, and it may be also related to the production of free radicals. The common deletion is often used as a spesific indicator of oxidative damage as well. Rosiglitazone is a drug which belongs to tiazolidinedione class used in the treatment of Type II diabetes mellitus. Drugs of this class acts as ligands for the peroxisome proliferators activated receptor-gamma.The aim of this study was to investigate the effects of rosiglitazone on the mitochondrial DNA by examining the frequency of the common deletion in the different rat tissues. For this purpose, nuclear and mitochondrial DNA was isolated from of liver, small intestine, pancreas and lymphocytes of the male Sprague-Dawley rats. Nuclear DNA, mitochondrial DNA and common deletion copy numbers of the samples was determined by quantitative PCR. The mitochondrial DNA copy number per cell and common deletion frequencies were calculated.The common deletion frequency in the liver tissue at all doses of rosiglitazone was found to be significiantly lower than in the control group. However, the mitochondrial DNA copy numbers per cell in the tissues of small intestine and pancreas were significiantly lower than in all rosiglitazone doses compared to the control group. At the same time, there was a significiant statistical difference in the common deletion frequency and mitochondrial DNA copy numbers per cell between the control groups.As a conclusion, rosiglitazone treatment let to decrease the common deletion frequency in the tissue of liver, indicating no oxidant effect of it. But rosiglitazone caused to decrease the mitochondrial DNA copy numbers in the tissues of small intestine and pancreas, this condition might be considered rosiglitazone?s oxidant effect.Key Words: Rosiglitazone, Common Deletion, Oxidative Stress
