Abstract
Bu çalışmada, A. auricula ve T. versicolor mantar türlerine ait etanol özütlerinin antioksidan, antimikrobiyal, DNA koruyucu aktivitelerinin ve mineral madde içeriğinin belirlenmesi amaçlanmıştır. Antioksidan aktiviteleri Rel Assay Diagnostics kitleri (TAS, TOS) ve DPPH yöntemi ile yapılmıştır. Antimikrobiyal aktivite testleri 5 farklı bakteri türü kullanılmış ve minimum inhibisyon metodu (MIC) ile çalışılmıştır. Mantar özütlerinin, DNA'yı koruma etkinliklerinin tespiti için pBR322 plazmid DNA'sı (vivantis) kullanılmıştır. Plazmid DNA, mantar örneklerinin varlığında hidrojen peroksit(H2O2) ve UV 'ye maruz bırakılarak hasara uğratıldıktan sonra. % 1,25'lik agaroz jel üzerinde görüntüleme gerçekleştirilmiştir. Mantarların mineral madde içeriği yaş yakma yöntemiyle mineralize edilip atomik absorbsiyon spektrofotometresi kullanılarak belirlenmiştir. Elde edilen sonuçlara göre mantar türlerinden elde edilen özütlerin, hem DPPH yöntemiyle hem de Rel Assay Diagnostics kitleriyle yapılan çalışmada önemli derecede serbest radikal temizleme aktivitesi göstermediği gözlenmiştir. Antimikrobiyal testler değerlendirildiğinde A. auricula özütünün test edilen bakteriler üzerine T. versicolor özütüne göre daha fazla bakteri üzerinde etkili olduğu belirlenmiştir. Yapılan DNA koruyucu aktivite testlerinde uygulanan konsantrasyonların pBR322 plazmid DNA üzerine etkisinin bulunmadığı tespit edilmiştir. In this study, we aimed that determine antioxidant, antimicrobial, DNA protective activity and mineral substance content of A. auricula and T. versicolor fungus species with ethanol extract. Rel Assay Diagnostics kits and DPPH method were used (TAS, TOS) to determination of antioxidant activity. For the test of antimicrobial activity 5 different bacteria species (Enterocossus faecalis 29213, Klebsiella pneumoniae 700603, Pseudomonas aeruginosa 27853, Escherichia coli 35218 and Staphyloccus aureus 28213) were used and studied with minimum inhibition method (MIC). pBR322 plasmid DNA was used UV-C and from induced oxidative damage to determination of protective activity of mushroom extracts. After, plasmid DNA and mushroom extracts damaged by hydrogen peroxide and UV, on 1.25% agarose gel imaging will be performed. Mineral matter content of fungi mineralized whether wet digestion method was determined using atomic absorption spectrometry. According to the results, not only DPPH but also Rel Assay Diagnostics kits didn't clean so much free radicals in the extract of A. auricula and T. vericolor. When evaulated antimicrobial tests, the extract of A. auricula more effective than extract of T. versicolor about the bacteria of study. Also the test of DNA protect activity didn't effect pBR322 plazmid, which tried concentration.
