Ozonlanmış fındık yağı`nın sitotoksik etki potansiyelinin belirlenmesi

Abstract
Fındık ülkemizde üretilen besleyici değeri yüksek olan son derece önemli bir tarım ürünüdür. Fındık yağının kullanımı da giderek artmakla beraber ozonlanmış formu daha önce hiç elde edilmemiştir. Ozonlu bitkisel yağlar, uygulanması ve elde edilmesi kolay olduğu için ozon tedavisinde çok tercih edilmektedir. Bu çalışma kapsamında 7-8 debide farklı sürelerde ozonlanarak elde edilen ve dansitesi, viskozitesi, peroksid ve iyot değerleri belirlenmiş olan fındık yağlarının sitotoksik etki potansiyelin belirlenmesi amacıyla bu çalışmada kontrol grubu olarak ticari form olan rafine fındık yağı kullanılmıştır. Ozonlanmış fındık yağlarının (2, 3, 4, 5 nolu örnekler) güvenli bir şekilde kullanılabilirliğinin tespiti için 3 farklı hücre hattında MTT testi uygulanması ile sitotoksik etki potansiyeli belirlendi. Bunun için H1299 akciğer kanseri, HUVEC endotel hücreleri ve A549 hücre dizisinde 48 ve 72 saat inkübasyon sonrası her numunenin 5 farklı konsantrasyonunda ki canlılık oranları hesaplandı. Sonuç olarak 3 farklı hücre hattına 5 farklı konsantrasyonda (100/50/25/12,5/6,25 µg/ml) uygulanan fındık yağlarının sitotoksik bir etkisinin olmadığı saptandı. Aynı zamanda rafine ozonlanmış fındık yağı ile kıyaslandığında numunelerin ozonlanma ile oluşan toksik bir etkisinin olmadığı da ortaya konuldu. Bu nedenle ozonlanmış fındık yağlarının, rafine fındık yağına alternatif bir ürün olarak ozonlama işleminden kaynaklanan herhangi bir toksik etkisinin olmadan kullanılabileceği bu çalışma ile ortaya konuldu. Bu bilgiler ışığında fındık yağının ozonlanmasının sitotoksik bir etkisinin olmaması güvenilir bir şekilde kullanılabileceği aynı zamanda ozonun gerek farmakolojik gerekse de kozmetik etkilerini de bünyesinde bulunduran fonksiyonel bir yağ olarak üretilebileceği kanaatine varılmıştır. Hazelnut is an extremely important agricultural product with high nutritional value produced in our country. The use of hazelnut oil has also increased and the ozonated form has never been obtained before. Ozonated vegetable oils are preferred for ozone treatment because they are easy to apply and obtain. In this study, refined hazelnut oil was used as a control group in this study in order to determine the cytotoxic effect potential of hazelnut oils obtained by ozonation at 7-8 different times and the density, viscosity, peroxide and iodine values were determined. Potential cytotoxic effect was determined by MTT test in 3 different cell lines to determine the safe use of ozonated hazelnut oil (samples 2, 3, 4, 5). For this, viability rates were calculated at 5 different concentrations of each sample after 48 and 72 hours incubation in H1299 lung cancer, HUVEC endothelial cells and A549 cell line. As a result, it was determined that there was no cytotoxic effect of hazelnut oil applied at 3 different cell lines at 5 different concentrations (100/ 50/ 25/ 12,5/ 6,25 μg/ml). At the same time, it was also shown that there was no toxic effect of ozonation of the samples when compared with refined ozonated hazelnut oil. For this reason, ozonated hazelnut oils can be used as an alternative to refined hazelnut oil without any toxic effects resulting from the ozonation process. This information has led to the conclusion that ozone can be used safely in the absence of a cytotoxic effect of hazelnut oil ozonation in the light, but it can also be produced as a functional oil containing pharmacological effects as well as cosmetic effects.
