LMP2 ve LMP7 genlerinin obezite ile ı̇lişkisinin ı̇ncelenmesi

Abstract
Obezite vücutta aşırı yağ birikimi sonucu oluşan ve kardiyovasküler hastalıklar, tip 2 diyabet ve kanser gibi hastalıkların riskini arttıran bir durumdur. Dünya Sağlık Örgütü tarafından yapılan sınıflandırmaya göre; vücut kitle indeksi 30'un üzerinde olan bir birey obez olarak tanımlanmaktadır. Çevresel faktörlerin yanısıra genetiğin de obezite gelişiminde etkili olduğu bilinmektedir. Çok sayıda ilişkili gen tanımlanmış olmasına rağmen, obezitenin genetik temeli henüz net olarak tanımlanmamıştır. Bu çalışmada, LMP2 ve LMP7 gen polimorfizmlerinin obezite gelişimindeki rollerinin araştırılması amaçlanmıştır. Morbid obez olarak tanımlanmış 100 birey ve normal vücut kitle indeksine sahip 100 kontrol bireyde LMP2-60 ve LMP7-145 polimorfizmleri PCR- RFLP tekniği ile analiz edilmiştir. Morbid obez ve kontrol grubunda allel ve genotip frekansları hesaplanmış ve karşılaştırılmıştır. Sonuç olarak, LMP2-60 ve LMP7-145 polimorfizmleri ile obezite arasında istatistiksel olarak anlamlı bir ilişki saptanamamıştır (p>0,05). Bu sonuçlara göre, LMP genlerinin obezite gelişiminde etkili olmadığı ifade edilebilir. Obesity is a situation resulting from excess body fat accumulation and increasing the risk of the dieases such as cardiovascular disorders, type 2 diabetes and cancer. According to the classification done by the World Health Organisation, an individual having body mass index higher than 30 is defined as obese. It is known that as well as environmental factors, genetics also has an effect in the development of obesity. Although many related genes have been reported, the genetic basis of obesity has not been clearly identified yet. The aim of the present study was to investigate the role of LMP2 and LMP7 gene polymorphisms in the development of obesity. LMP2-60 and LMP7-145 polymorphisms were analysed by PCR-RFLP technique in 100 morbid obese patient and 100 control individuals having normal body mass index. The allele and genotype frequencies were calculated and compared between morbid obese and the control group. As a result, no statistically significant association was found between LMP2-60 and LMP7-145 polymorphisms and obesity (p>0.05). According to these result, it can be concluded that these genes do not have a role in the development of obesity.
