Abstract
Sınırlı enerji rezervleri ve fosil yakıtlardan dolayı atmosferde artan sera gazları nedeniyle, biyodizel (Yağ asidi metil esterleri, YAME), her ülkenin politik gündeminin bir konusu haline gelmiştir. Bu çalışmada serbest Thermomyces lanuginosa lipazı üre hidrolizi yöntemi ile hazırlanan Mg-Al hidrotalsite ve inorganik perlite tutuklanmıştır. Tutuklanmış lipaz katalizörlüğünde atık yemeklik yağın metanol ile transesterleşmesiyle biyodizel üretimi gerçekleştirilmiştir. Çalkalama hızı (150 ? 300 rpm), sıcaklık (25 ? 45 °C), başlangıç su miktarı (%0-20), metanol ekleme sıklığı (tek, iki ve üç basamak), enzim miktarı (%1-10), yağ/metanol mol oranı (1/3, 1/4, 1/6), çözücü türü (hekzan, isooktan, propanol, toluen) gibi parametrelerin biyodizel üretimine etkileri incelenmiştir. Mg-Al hidrotalsite tutuklanmış lipaz Thermomyces lanuginosa serbest lipazının, sıvı formdaki Lipozyme TL-100 L ve ticari tutuklanmış formu Lipozyme TL-IM?nin transesterleşme reaksiyonundaki performansları karşılaştırılmıştır. Reaksiyonlar 6ml?lik viallerde 24 saatlik sürede gerçekleştirilmiştir. Deneysel çalışmalar sonucunda, Mg-Al hidrotalsit için tutuklama verimi %36, inorganik perlit için %15 olmuştur. Transesterleşme reaksiyonu deneyleri sonucunda optimum su içeriği sırasıyla Lipozyme TL-100 L, Lipozyme TL-IM ve UHT-TL için %10, %15, %1 olarak bulunmuştur. 24 saat sonunda her enzim için optimum su içeriğinde, 35°C sıcaklıkta, çalkalama hızı 200 rpm, enzim miktarı %5, yağ/metanol mol oranı 1/4 ve metanol 3 basamak eklendiği koşulda en yüksek YAME içeriği üç enzim için sırasıyla %98, %83 ve %92 olarak bulunmuştur. Due to limited energy reserves and the increasing environmental pressure on green house gases coming from the fossil fuels, biodiesel (Fatty acid methyl esters, FAMEs), is becoming the hot topic of every country's policy agenda. In this study, Thermomyces lanuginosus lipase was immobilized on Mg-Al hydrotalcite prepared by the hydrolysis of urea and inorganic perlite. Biodiesel production of waste cooking oil with methanol by lipase catalyzed was investigated. Agitation speed (150?300 rpm), temperature (25?45 °C), initial water content (%0-20), methanol addition frequency (one, two, three step), amount of enzyme (%1-10), oil/methanol molar ratio (1/3, 1/4, 1/6), solvent type (hexane, isooctane, propanol, toluene) the effects of parametres on biodiesel reaction were studied. Transesterification reaction performances of commercially available immobilized lipase (Lipozyme TL-IM) and free lipase (Lipozyme TL 100 L) were compared to the immobilized lipase on the Mg-Al hydrotalcite (UHT-TL). The reactions were carried out in the 6ml volumed vials for 24 hours. According to the experimantal studies immobilization yield for Mg-Al hydrotalcite, inorganic perlite 36%, 15% respectively. At the end of transesterification reaction experiments it was found that initial water content of the media for UHT-TL, Lipozyme TL-IM and Lipozyme TL 100 L %10, %15, %10 respectively. When the reaction time of 24 hours in 1/4 oil/alcohol molar ratio, 3 step methanol addition at 35 °C with 5% the amount of enzyme, 200 rpm agitation speed and 1%, 15% and 10% water contents, the yields of FAME were 98%, 83%, and 92%.
