Candida albicans üzerine kalsiyum hidroksit, klorheksidin ve propolis`in antimikrobiyal etkinliğinin kıyaslanması

Abstract
Bu çalışmanın amacı propolis, kalsiyum hidroksit, klorheksidin, kalsiyum hidroksit +klorheksidin karışımı ve kalsiyum hidroksit + potasyum iyodür karışımlarının iki farklı zaman diliminde C. albicans üzerine antimikrobiyal etkinliklerini araştırmaktır.Toplam 120 çekilmiş insan kesici dişi 12 mm kök kalacak şekilde mine-sement sınırından kesildi. Kök kanalları NiTi döner aletlerle, #40/04' e kadar genişletildi. Preparasyon ve sterilizasyon işlemlerinden sonra örnekler C. albicans ile kontamine edildi. Yirmi bir günlük inkübasyonun ardından, kanal içi ilaçlama metoduna göre, örnekler altı gruba ayrıldı. Grup 1: propolis, Grup 2: Ca(OH)2, Grup 3: %2'lik CHX jel, Group 4: Ca(OH)2 + %2'lik CHX, Grup 5: Ca(OH)2 + potasyum iyodür, Grup 6: steril salin (pozitif kontrol'dir). Canlı kalan mantar populasyonunu hesaplamak üzere koloni sayımı; i) birinci günün sonunda (her grubun yarısı), ii) yedinci günde (grupların diğer yarısı) yapıldı. Verilerin istatiksel değerlendirmesi için Kruskal Wallis testi, Mann Whitney-U testi uygulandı. Birinci günün sonunda, tüm gruplar, kontrol grubuna göre istatiksel olarak anlamlı farklılık gösterdi (p < 0.05). Klorheksidinin tek ya da kalsiyum hidroksitle kombine kullanımı en yüksek antifungal etkiyi gösterdi.Yedinci günde, tüm çalışma grupları, ilk günün sonundaki durumlarına kıyasla belirgin düzeyde daha fazla antifungal etki gösterdi. Klorheksidinin tek başına ya da kalsiyum hidroksitle kombine kullanımı C. albicans eliminasyonunda etkili bulundu. Propolis'in antifungal etkisi görülmektedir fakat diğer gruplardan istatistiksel olarak anlamlı bir farklılık bulunmamıştır.Anahtar Kelimeler: Propolis, Candida albicans, Klorheksidin The present study aimed to investigate the antifungal activity of propolis, calcium hydroxide, 2% chlorhexidine, calcium hydroxide with chlorhexidine and calcium hydroxide with iodine potassium iodide on Candida albicans-infected root canal dentinal tubules at two time intervals. A total of 120 extracted human anterior teeth were sectioned below the cementoenamel junction to obtain 12 mm of the roots. Root canals were enlarged with rotary files up to #40/04. Following the preparation and sterilization, the specimens were contaminated with C. albicans. The samples were incubated for 21 days. Then, the specimens were divided into six groups based on intracanal medication. Group 1: propolis, Group 2: Ca(OH)2, Group 3: 2% chlorhexidine gel, Group 4: Ca(OH)2 with 2% CHX, Group 5: Ca(OH)2 with iodine potassium iodide, Group 6: sterile saline as positive control. The number of colony forming units were calculated for assessing the remaining vital viable fungal population; i) at the end of the 1st day (half of each group), ii) at the 7th day (the other halves). The values were analyzed statistically using non paramatric Kruskal-Wallis and Mann-Whitney-U tests to compare the median reduction of C. albicans. At the first day, the differences among the groups were statistically significant compared to the control group ( p<0.05). Ca(OH)2 with CHX and CHX alone were the most effective against C. albicans. At day 7, all medicaments presented further significant antifungal effect compared to their first day counterparts. Treatments with CHX alone or CHX combination with Ca(OH)2 were effective for the elimination of C. albicans. Propolis had apparent antifungal effect, however its effectivness was generally not superior to those of the other tested medicaments.Keywords: Propolis, Candida albicans, chlorhexidine.
