Elazığ yöresinde yetişen dut, kızılcık, kiraz ve ceviz meyvelerinin antioksidan kapasiteleri ve bazı deney modellerinde oluşturulan oksidatif stres üzerine etkilerinin incelenmesi

Abstract
Bu çalışmada, beslenme açısından yaygın bir kullanım alanına sahip olan, dut, ceviz, kiraz ve kızılcık meyvelerinde bazı fitokimyasal analizler yapıldı ve bu meyvelerin maya hücresi ile sıçan dokularında H2O2 ve CCl4'ün toksititesine karşı biyolojik etkileri araştırıldı. Analiz sonuçlarına göre, flavonoid içeriği en yüksek olan meyvenin karadut, lipofilik vitaminler ve fitosterol içeriği en fazla olan meyvenin ise kızılcık olduğu belirlendi. Şeker içeriğinin ise dut meyvelerinde daha yüksek olduğu tespit edildi. İn vitro deney sonuçlarına göre, standart Saccharomyces cerevisiae kültürüne H2O2 ilave edilmesi sonucu palmitik (16:0), stearik (18:0), palmitoleik (16:1, n-7) ve oleik (18:1, n-9) asitlerin düzeylerinde artışlar gözlendi. Meyvelerin kullanıldığı kültürlerde ise bu yağ asitlerine ilaveten linoleik (18:2, n-6) ile linolenik (18:3, n-3) asit düzeylerinde de artışlar belirlendi (p<0.05, p<0.001). S. cerevisiae kültürüne CCl4, rutin ve flavonoid ekstraktının ilave edilmesi sonucu bazı yağ asitleri, ?-tokoferol, ergosterol ve K vitamini miktarlarında artış olduğu tespit edildi (p<0.05).Sıçanlar üzerinde yapılan çalışmada, CCl4 uygulaması sonucu, serum ve diğer dokulardaki bazı yağ asidi düzeylerinin yükseldiği saptandı (p<0.001). CCl4'e karşı ceviz ekstraktı ve rutin uygulaması yapılan grupların serumdaki yağ asidi düzeylerinin kontrol grubu değerlerine yakın olduğu gözlendi. H2O2 ve CCl4 uygulamasına karşı karadut ve ceviz ekstraktı verilen gruplarda, bazı dokularda GSH düzeyinin yükseldiği ve MDA düzeyinin de azaldığı belirlendi (p<0.05, p<0.01). Lipofilik vitamin düzeyleri incelendiğinde, eritrositlerdeki ?-tokoferol düzeyi rutin+CCl4 grubu dışında azalmasına rağmen, diğer dokularda artışlar gözlendi (p<0.05). Kolesterol düzeyinin de serum ve dokularda kontrol grubuna göre yükseldiği tespit edildi (p<0.05, p<0.001).Sonuç olarak, in vitro ortamda meyve, CCl4 ve H2O2 ile yapılan çalışmalarda yağ asidi, vitamin ve fitosterol sentezinde kullanılan enzimlere ait son ürünlerin miktarlarında değişik oranlarda farklılıklar gözlendi. Benzer sonuçların sıçan dokularındaki yağ asidi ve kolesterol sentezinde de gerçekleştiği saptandı.Anahtar Kelimeler: Ceviz, dut, GSH, karadut, kızılcık, kiraz, kolesterol, lipofilik vitaminler, MDA, Saccharomyces cerevisiae, sıçan ve dokular, yağ asidi. In this study, in terms of nutrition, some phytochemical analysis were performed in mulberry, walnut, cherry and cranberry fruits which have a widespread use, and biological effects of these fruits on H2O2 and CCl4 against toxicity in yeast cells and rat tissues were investigated. According to the results, the highest flavonoid content has been found in black mulberry, and the highest lipophilic vitamins and pyhtosterol content has been in cranberry. Mulberry fruits were found to be higher in sugar content. In the test results of In vitro, when H2O2 was added to the standard Saccharomyces cerevisiae culture, the increase in palmitic (16:0), stearic (18:0), palmitoleic (16:1, n-7) and oleic (18:1, n-9) acids were observed. In the cultures where fruits has been used, in addition to these fatty acids, linoleic (18:2, n-6) and linolenic (18:3, n-3) acid levels were also detected (p<0.05, p<0.001). When CCl4, rutine and flavonoid extract added to S. cerevisiae culture, an increase in the levels of some fatty acids, ?-tocoferol, ergosterol and vitamin K were observed (p<0.05).The study on rats, as a result of CCl4 application, an increase in some of the fatty acid levels were observed in serum and other tissues (p<0.001). Against the CCl4, the groups using walnut extract and rutine application were close to the values of the control group in terms of fatty acid level in serum. Against the H2O2 and CCl4 applications, GSH level was increased and MDA level decreased in some tissues of rats when mulberry and walnut extract applied (p<0.05, p<0.01). Although ?-tocoferol levels in erythrocytes decreased with the exception of the rutine+CCl4 group an increase was observed in other tissues (p<0.05). The levels of cholesterol in serum and tissues of applied groups were higher than the control group (p<0.05, p<0.001).As a result, in the studies with fruit, CCl4 and H2O2 in in vitro media, differences were observed in different proportions in the amounts of the latest products of enzymes that were used in the synthesis of fatty acid, phytosterol and vitamin.
