Abstract
Bu çalışmada kullanılmak üzere, çeşitli toprak ve evsel atık su örneklerinden, üre agar kullanılarak; üreaz enzimi üreten çok sayıda bakteri türü izole edilmişi bunlar arasından 6 adet Proteus vulgaris straini tanımlanmıştır. Doğadan izole edilen 6 Proteus vulgaris straini arasından üreaz verimi en yüksek olan strain, fermentasyon yoluyla seçilmiştir. üreaz verimi en yüksek olan strain kullanılarak, onun üreaz biyosentezi ve aktivitesine etkili olan bazı faktörler saptanmıştır. üreaz biyosentezi üzerinde değişik sıcaklık, pH inkübasyon süresi ve inokulum miktarının etkileri araştırılmış, 25°C 'de. 7.5 pH'da. 3 ml/100 mi inokulum ve 8 saatlik inkübasyon süresi kullanıldığında en yüksek üreaz verimi saptanmiştır. Son olarak, yüksek üreaz aktivitesi veren optimum koşullar araştırılmış ve 25 *C 'de, 7. 5 pH'da. fcl'lik üre konsantrasyonunda ve 10 dakikalık bir inkübasyon süresinde yüksek üreaz aktivitesi elde edilmiştir. üreaz enzimi aktivitesi saptanmasında nesslerizasyon yöntemi kullanılmıştır. In order to use in this study, from the samples of various soils and domestic waste waters, by using urea agar, a number of ureolytic bacteria were isolated and 6 strains of Proteus vulgaris were identified among these. The strain which gives the highest yield of urease was selected among 6 Proteus vulgaris strains by using fermentation technique. By using the one' which gives the highest yield of urease, some physiological factors were determined on its urease biyosenthesis and its urease activity. The effedts of various temperatures, pH, duration of incubation and the amounts of inokulum were studied and the highest urease biyosenthesis were determined at the temperature of 25*C, at pH 7.5, in 3ml/100 ml of inoculum and in 8 hours of incubation duration. Finally, the optimum conditions on highest urease activity were determined at the ternparature 25 *C, at pH 7.5, in the %1 urea concentration and 10 minutes duration. The nesslerization method was used on the determination of urease activity.
