Abstract
ÖZET Galleria mellonella (LEPİDOPTERA)'NIN SPERM KESELERİNDE FERTİL VE STERİL SPERMLERİN İNCE YAPI FARKLARI VE GLİKOZAMİNOGLİKANLARI SARIBEK, Büget Yüksek Lisans Tezi, Biyoloji Anabilim Dalı Tez Yöneticisi: Yrd.Doc.Dr. Remziye DEVECİ Kasım 2002, 57 sayfa Bu çalışmada, G.mellonella'da üretilen fertil ve steril spermler arasındaki yapısal farklar, ışık ve elektron mikroskobu ile incelenmiştir. Ayrıca, bu iki sperm tipi arasında glikozaminoglikanlar açısından farklılıklar olup olmadığı da farklı konsantrasyonlarda MgCl2 içeren Alcian blue ile histokimyasal boyamalar yapılarak araştırılmıştır. Testisteki fertil ve steril spermatidler ayrı demetler oluştururlar. Fertil spermatidlerin nukleusları demetin anteriorunda, aynı hizada görülür. Steril spermatidlerde ise nukleuslar nukleuslar sperm demeti boyunca dağınıktır. Fertil spermler nukleuslarının uzaması, steril spermler ise nukleuslarını kaybederek testisteki gelişimini tamamlar. Sperm kesesine geçen olgun fertil spermlerin silindirik nukleusları demetin anteriorunda birlikte koyu bir bant oluşturur. Sperm kesesindeki steril spermlerde ise nukleus bulunmaz ve spermler dağınık görülür. Olgun fertil ve steril spermlerde mikrotübüllerden oluşan bir aksonem,VI kuyruk boyunca uzanır. Olgun fertil spermlerde homojen yapıya sahip iri bir mitokondriyum ayırt edilir. Olgun steril spermlerde ise eşit büyüklükte iki mitokondriyum vardır. Farklı konsantrasyonlarda MgCl2 içeren Alcian blue boyamaları ile fertil ve steril spermlerde glikozaminoglikanların varlığı belirlenmiştir. Bunlardan karboksilli olanlar, sülfatlılara oranla daha boldur. Sülfatlı glikozaminoglikanlardan sadece kondroitin sülfat ve dermatan sülfat, fertil spermlerin baş bölgesinde çok az oranda bulunur, kuyruk bölgesinde ve steril spermlerin tamamında yoktur. Also, whether there are differences between the two types of sperms in content of glycosaminoglycans was investigated by using histochemical stains by Alcian blue including MgCl2 in different concentrations. Fertile and sterile spermatids in the testis forms separate bundles. The nuclei of fertile spermatids are seen in same line, in anterior of the sperm bundles. In sterile spermatids, on the other hand, nuclei are scattered through the sperm bundles. Fertile sperms complete then- development by elongating their nuclei while sterile sperms complete their development by losing their nuclei. The together cylindric nuclei of mature fertile sperms which passes to vesicula seminalis form a dark band in the anterior of the bundle. There is no nucleus in sterile sperms invııı the vesicula seminalis and sperms are scattered. In mature fertile and sterile sperms, an axoneme composed of microtubules lies through the tail. In mature fertile sperms, a large mitochondrial derivative which has homogenous structure is distinguished. In mature sterile sperms, on the other hand, are two mitochondrial derivatives which have the equal size. The presence of glycosaminoglycans in fertile and sterile sperms was determined by Alcian blue stains containing MgCl2 in different concentrations. From these, those are carboxylated are more abundant than sulphated ones. Of the sulphated glycosaminoglycans, only chondroitin sulphate and dermatan sulphate are present in less ratio in head region of fertile sperms, there aren't in tail region of fertile sperms and any of them in sterile sperms.
