Candida albicans (Robin) Berkhout`un species-specific PCR ile DNA düzeyinde tanısının standardizasyonu

Abstract
Bu çalışmada C. albicans 'in Species-Specific PCR ile tanısının yapılması amaçlanmıştır. Bu amaç ile 5 C.albicans referans straini, 9 farklı Candida türü (referans strainler ve klinik izolatlar), Cyrptococcus neoformans, 4 filamentöz fungus (referans strainler ve klinik izolatlar), 3 bakteri referans straininden DNA izolasyonu yapılmıştır. İnsan DNA 'sı hazır olarak elde edilmiştir.Ca/fo'cörcs,'ın büyük alt birim rDNA gen bölgesinde yer alan D1/D2 bölgesine spesifik primer seti (CAL5,NL4CAL,NL5CAL) kullanılmış ve Species-Specific PCR'ın standardizasyonu yapılmıştır. Bu amaç ile farklı MgCİ2 konsantrasyonları, agaroz konsantrasyonları, farklı PCR döngü profilleri, farklı DNA konsantrasyonlarının PCR'a etkisi denenmiştir. Bu primerlerin 19 pg kromozomal DNA' yi saptadığı bulunmuştur. Bu PCR 'da bu primerler sadece C.albicans kromosomal DNA'sı mevcut iken 175 bç'lik (baz çifti)'lik ürün oluşturmuştur ve fenotipik olarak C.dubliniensis ve C.tropicalis olarak tanılanmış iki klinik izolat hariç diğer DNA 'lar mevcut olduğunda hiçbir ürün oluşmamıştır. C.albicans DNA' sim içeren ve 2 farklı Candida (CDU2,NL4CAL ve CW02,L4LEL) türüne ait primerler ile bir seri PCR gerçekleştirilmiş ve hiçbir çoğaltım gözlenmemiştir.Anahtar Kelimeler: C.albicans, DNA, Standardizasyon, Species-Specific PCR In this study, we aimed identification of C.albicans by Species- Specific PCR chromosomal DNA of 5 reference strains of Candida albicans, 9 different Candida species (clinical isolates and reference strains), Cyrptococcus neoformans (clinical isolate), 4 micelial fungi (clinical isolate and reference strains), 3 reference strains of bacteria was extracted and purified. And human DNA was available. We used primer set (CAL5,NL4CAL,NL5CAL) for the D1/D2 region in the large-subunit rDNA gene of C.albicans and standardization of Species- Specific PCR. For this aim different MgC^and agarose concentration, different profile of PCR cycle was tested. The detection limit of these primers was found 19 pg chromosomal DNA. In this PCR, these primers yielded DNA fragments of 175 bp (base pair) when the C.albicans DNA was present and didn't yield any product with the any other DNA except C.dubliniensis (clinical isolate) and C.tropicalis (clinical isolate) which were identified phenotypically. Furthermore, series PCRs with two different primer pairs are specific two different Candida species (CDU2,NL4CAL and CW02,L4LEL) contained chromosomal DNA of C.albicans not yield any product.
