Galleria mellonella (Lepidoptera)`da lektinler ve analitik yöntemlerle sialik asit (Neu5Ac) belirlenmesi

Abstract
Bu çalışmada Galleria mellonella (Lepidoptera)'nın yağ dokusunda Sialik asit (Sia)'ın varlığı metamorfoz sırasında (sondan bir önceki ve son larval devreler ile pupa ve ergin devrelerde) fluoresan mikroskobu ve analitik ESI-MS/MS yöntemleri ile belirlenmiştir.Fluoresan işaretlemeler için Fluorescein isothiocyanate (FITC) işaretli lektinler uygulanmıştır. Tüm Sia tipleri ile bağlanan LFA (Limax flavus agglutinin) ve LPA (Limulus polyphemus agglutinin) lektinleri ile Neu5Ac?2,3Galß1,4GlcNAc dizisini tanıyan MAA (Maackia amurensis agglutinin) ve Neu5Ac?2,6Gal/GalNAc dizisini tanıyan SNA (Sambucus nigra agglutinin) lektinleri kullanılmıştır.ESI-MS/MS belirlemeleri için önce standart Sia (N-Acetylneuraminic Acid-NANA, Sigma A-9646, FW;309.3) MS/MS'e infüzyon pompasından örnek akış oranı 250 µl/h olacak şekilde verilmiş ve parçalanma ürünleri belirlenmiştir. Disekte edilerek metanolde biriktirilmiş doku örnekleri asidik (4M asetik asit) hidroliz sonrasında liyofilize edilerek saklanmıştır. Metanolde çözülen örnekler aynı akış oranı ile ESI-MS/MS sistemine verilmiştir. Doku örneklerinde de kontroldaki Sia parçalanma ürünleri aranmıştır.Fluoresan mikroskobu ön incelemeleri, frozen mikrotomla kesilmiş ve Hematoksilen-Eosin boyasıyla boyanmış kesitlerde yapılmıştır. uygulama sonuçlarına göre Sia incelenen bütün devrelerde bulunmaktadır. ?2,3 bağlı Neu5Ac?2,3Galß1,4GlcNAc için özel olan MAA ışıması pupal dönemde zayıf diğer dönemlerde daha güçlüdür. ?2,6 bağlı Neu5Ac?2,6Gal/GalNAc dizisini tanıyan SNA ışıması sondan bir önceki larva ile ergin dönemlerde daha güçlü son larva ve pupal dönemlerde daha zayıf bulunmuştur.ESI-MS/MS sonuçlarına göre standart örneğin parçalanma ürünleri olan 290, 220, 170, 119 ve 87 değerleri örneklerin çoğunda bulunmuştur. 290m/z bütün örneklerde en belirgin bulunurken 220, 170 ve 119 değerleri örneklerin çoğunda küçük değerlerde de olsa görülmüştür. En son parçalanma ürünü olan 87m/z değeri metamorfozun ileri devrelerinde daha belirgin bulunmuştur.Sonuç olarak fluoresan mikroskobu ve ESI-MS/MS sonuçlari G.mellonella'nın metamorfoz sırasında incelenen devrelerin yağ dokularında Sia (?2,3 ve ?2,6 bağlı Neu5Ac) bulunduğunu göstermektedir. Elektron mikroskobu incelemelerinde katyonik boya Rutenyum Kırmızısı ile oluşan pozitif reaksiyonun bazı yağ damlalarında görülmesi, negatif yüklerce zengin Sia'nın buralarda bulunabilecini işaret etmektedir.Anahtar Kelimeler: Sialik asit, Lektin işaretleme, Fluoresan mikroskobu, Rutenyum kırmızısı, Elektron Mikroskobu, Mass Spektrometri, Yağ dokusu, Galleria mellonella, Böcek In this study, the presence of Sialic acids in fat body of Galleria mellonella(Lepidoptera) during metamorphosis (penultimate and last larval stages, pupae and adult stages) was determined that by a combination of fluorescence microscopy and analytic ESI-MS/MS technique.For fluorescence labeling, Fluorescein isothiocyanate (FITC) labeled lectins was used. LFA (Limax flavusagglutinin) and LPA (Limulus polyphemus agglutinin) lectins, which bind to all Sia types, and also MAA (Maackia amurensis agglutinin) and SNA (Sambucus nigraagglutinin), which respectively recognize Neu5Ac?2, 3Galß1, 4GlcNAc chains and Neu5Ac?2,6Gal/GalNAc chains,.Initially, standard Sia (N-Acetylneuraminic Acid-NANA, Sigma A-9646, FW; 309.3) were applied to MS/MS by infusion pump with the flow rate of 250 µl/h and the daughter ions of Sia was determined. Then, fat body samples were dissected from the insect and collected into methanol. After that acidic hydrolysis (4M acetic acid) was performed on the collected material and then materials were lyophilized. Finally, samples were dissolved into methanol and applied to ESI-MS/MS with the same flow rate. The samples were then analyzed for the presence of daughter ions with standard Sia.The sample sections that were obtained by frozen microtome and stained with Hematoxylen-Eosin were used for pre-investigation followed by fluorescence microscopy. According to the results obtained, Sias were present in allstages of development. The MAA radiance that is specific for ?2,3 which binds to Neu5Ac?2,3Galß1,4GlcNAc was lower in pupal stage, whereas it was higher other stages. The SNA radiance specific for Neu5Ac?2,6Gal/GalNAc was higher in penultimate larval and adult stages than last larval and pupal stages.The fragments of standard Sia, 290, 220, 170, 119, 87, were found in the most of the samples according to the ESI-MS/MS results. 290 m/z fragment was more distinct in all of samples compared to others, whereas 220, 170 and 119 fragments were less in most of samples. 87 m/z fragment ,which is the smallest fragment, were observed much more distinct in further stages of metamorphosis.As a result of this study, fluorescence microscopy and ESI-MS/MS findings indicated that Sias (?2,3 and ?2,6 binded Neu5Ac) are present in fat body of Galleria mellonella during the metamorphosis.
