?-galaktozidaz enziminin immobilizasyonu için biyoafinite temelli immobilizasyon prosedürlerinin geliştirilmesi

Abstract
?-Galaktozidazlar (?-D-galaktozid galaktohidrolaz, EC 3.2.1.22) hidrolazların bilinen bir sınıfı olup rafinoz ailesi şekerlerin galakto oligosakkaritleri ve galaktomannan polisakkaritlerdeki ?-1,6 bağlı D-galaktoz birimlerinin hidrolizini katalizlerler. Doğada oldukça yaygın olarak bitkilerde, hayvanlarda ve mikroorganizmalarda bulunurlar. ?- Galaktozidazlar çok çeşitli biyoteknolojik ve medikal uygulamalara sahiptir, örneğin; gıda işleme ve hayvan besleme prosedürlerinde, şeker endüstrisinde, kağıt hamuru ve kağıt endüstrisinde, enzimatik sentezlerde, yapısal analizlerde, kan grubu dönüşümlerinde ve Fabry hastalığının tedavisinde kullanılırlar.Enzim immobilizasyonu teknolojisi enzimlerin tekrar kullanılabilmesini sağlamak ve kararlılığını geliştirmek için etkin bir aracıdır. Enzimlerin immobilizasyonu için bazı protokoller mevcuttur ancak çok azı basit ve/veya enzim özelliklerini geliştirebilir niteliktedir. Bu nedenle yeni immobilizasyon protokollerine ihtiyaç duyulmaktadır. Enzimlerin immobilizasyonunda yeni trend enzim ve taşıyıcı arasında çeşitli biyoafinite bağlarının oluşturulmasıdır. Biyoafinite immobilizasyonunda enzim taşıyıcıda biyospesifik etkileşimlerle immobilize edilir.Bu çalışmada, ?-galaktozidaz enzimi domatesten (Lycopersicon esculentum) izole edildi ve sonra bir biyoafinite protokolü ile kitosan (doğal bir polimer) ve Sepabead EC-EA'da (sentetik bir polimer) immobilize edildi. Bu immobilizasyon protokolünde polimerler önce glutaraldehid ile aktive edildi ve ardından 3-aminofenil boronik asit (AFBA) ile türevlendirildi. Her iki biyoafinite taşıyıcısı ?-galaktozidaz enziminin immobilizasyonunda kullanıldı. İmmobilizasyon protokolü optimize edildi ve immobilizasyona bazı parametrelerin etkisi araştırıldı. Serbest ve immobilize enzimlerin fiziksel ve kimyasal karakterizasyonu gerçekleştirildi. Enzim aktivitesine etki eden bazı parametreler (pH, sıcaklık, substrat konsantrasyonu, efektör konsantrasyonları) incelenerek kararlılık testleri yapıldı. İmmobilize enzimlerin tekrar kullanılabilirliği de test edildi. Serbest ve immobilize ?-galaktozidaz enzimleri rafinoz hidroliz prosesinde kullanıldı. ?-Galactosidases (?-D-galactoside galactohydrolase, EC 3.2.1.22) are a common class of hydrolases that catalyze the hydrolysis of ?-1,6-linked-D-galactose residues present in galactooligosaccharides of sugars of the rafinose family and galactomannan polysaccharides. They are widely distributed in nature and found in plants, animals, microorganisms. ?-Galactosidases have many potential biotechnological and medicinal applications such as in food processing, animal feed prcocessing, sugar producing industry, pulp and paper industry, enzymatic synthesis, structural analysis, conversion of blood type and treatment of Fabry?s disease.Enzyme immobilization technology is an effective means to perform enzyme reuse and to improve its stability. There are many protocols for the immobilization of enzymes but very few are also very simple and/or very capable of improving enzyme properties. Therefore novel immobilization protocols are still needed. A new trend in enzyme immobilization is to create different bioaffinity bonds between the carrier and enzyme. In bioaffinity immobilization the enzyme is immobilized to a matrix via biospecific interactions.In this study, ?-galactosidase was isolated from tomato fruit (Lycopersicon esculentum) and then immobilized on chitosan (natural polymer) and Sepabead EC-EA (synthetic polymer) by using a bioaffinity protocol. In this protocol, the polymers activated by glutaraldehyde at first and then derivatized by 3-aminophenyl boronic acid (AFBA). Both the bioaffinity supports were used for the immobilization of ?-galactosidase. The immobilization protocol was optimized and several parameters that effect the immobilization of the enzyme were also investigated. Physical and chemical characterization of free and immobilized enzymes were made. Some parameters effecting to the enzyme activity (pH, temperature, substrate concentration and effector concentration) were searched and stability tests (thermal, pH, storage and operational stability) were also made. Beside of these, usebility of the immobilized enzymes were tested. Free and immobilized ?-galactosidases were also used in the hydrolysis of raffinose.
