Abstract
Levan, fruktoz ünitelerinin ß-2,6 bağlarıyla bağlanmasından oluşan bir fruktoz polimeridir. Bu çalışmada, Z. mobilis suşları ile sentetik ortam kullanılarak levan üretimi gerçekleştirilmiş ve en yüksek levan üreten suş olan Z. mobilis subspecies mobilis NRRL B-14023 ile levan biyosentezi üzerine başlangıç şeker konsantrasyonu, süre, sıcaklık, karıştırma-havalandırma, pH ve farklı azot kaynaklarının etkileri incelenmiş, bunlardan; başlangıç şeker konsantrasyonu (250-350g/L), fermentasyon süresi (24-48 saat) pH'nın (4.5-5.5) levan üretimine etkilerini incelemek ve bu parametrelerin optimum seviyelerini belirlemekamacıyla Cevap Yüzey Yöntemi (CYY) kullanılmıştır. Şeker konsantrasyonu, inkübasyon süresi ve pH'nın levan biyosentezi üzerine önemli bir etkiye sahip, regresyon modelinin önemli (P<0.05) ve modelin matematiksel uygunsuzluğundan kaynaklanan hatanın önemsiz (P>0.05) olduğu belirlenmiştir. Deneysel veriler kullanılarak bir matematik model oluşturulmuş ve bu verilerle modelden elde edilen verilerin uyumlu olduğu tespit edilmiştir. Bu model yardımıyla proses için optimum koşullar olan 299.11 g/L başlangıç şeker konsantrasyonunda, 42.34 saat inkübasyon süresinde ve pH 6'da, modelin tahminlediği değer 42.6 g/L iken üretim sonunda 40.2 g/L konsantrasyonunda levan elde edilmiştir. Zymomonas mobilis NRRL B-14023 hücreleri Ca-aljinat boncuklarında immobilize edilmiş ve dolgulu yatak biyoreaktörde sürekli sistemde levan üretim denemeleri gerçekleştirilmiştir. En yüksek levan üretimi, seyreltme hızının 0.14h-1 olduğu denemede 31.8 g/L olarak ve verimlilik 4.45 g L-1 h-1 olarak bulunmuştur. In this study, the production of levan from synthetic medium by Z. mobilis subspecies mobilis NRRL B-14023 and the effects of initial sugar concentration, fermentation time, temperature, agitation-aeration, pH and different nitrogen sources on levan production was investigated and Response Surface methodology was used to investigate the effects and optimum levels of three selected parameters; initial sugar concentration, (250-350 g/L ), fermentation time (24-48 hour) and pH (4.5-5.5) on levan production. Initial sugar concentration, fermentation time and pH had a strong lineareffect on levan concentration. Regression model was significant (P<0.05) and lack of fit was insignificant (P>0.05). Results of the statistical analysis showed that the fit of the model was good in all cases. Maximum levan concentration of 40.2 g/L was obtained when the predicted value was 42.6 g/L at the optimum levels of process variables (initial sugar concentration 299.11 g/L, fermentation time 42.34 hour, pH 6) . These values were obtained by fitting of the experimental data to the model equation. The production of levan by Zymomonas mobilis subspecies mobilis NRRL B-14023 immobilized in Ca-alginate beads in packed bed bioreactor was studied using continous fermentation system. The highest levan concentration (31.8 g/L)was obtained at a dilution rate of 0.14 h-1 with a productivity rate of 4.45 g L -1 h-1 and the highest productivity value (6.55 g L-1 h-1), was obtained at a dilution rate of 0.22 h-1 with a levan production rate of 29.8 g/L.Key words: Levan, microbial polysaccharide, Zymomonas mobilis, Optimization, İmmobilization, Levansucrase, Response Surface Methodology, Packed Bed Bioreactor
