dc.description.abstract | -27- VI. ÖZET Bu çalışmada mast hücreleriyle E vitamini arasındaki ilişki incelen di. Araştırmada i.p. 0.2 ml Complete Freunds Adjuvant enjeksiyonu ile immunize edilen 14 fare kullanıldı. îki gruba ayrılan deneklerden kontrol grubunda olanlara (7) 10 gün süreyle i.m. 0.002ml/gr SF enjekte edildi. Deney grubundakilere (7) i. m., 0.3 mg/gr vücut ağırlığı başına alfa tokoferol asetat (E vitamini) verildi. Çalışma sonunda kanda, periton sıvısından ve mezenterden alınan örneklerde histamin miktarları tayin edildi. Aynca periton sıvısı ve mezenter doku örneklerinde mast hücreleri toluidin mavisi ile boyanarak incelendi. Kontrol farelerin kanlarından ortalama histamin düzeyleri 1.6781 ± 0.4465 y/ml iken deney grubunda 0.2387 ± 0.0543 y/ml idi ve aradaki fark önemliydi (P < 0.01). Periton sıvısındaki histamin düzeylerinin ortalaması kontrol grubunda 1.8306 ± 0.3539 y/ml iken deney grubunda 0.2867 ± 0.0964 y/ml bulundu, fark önemliydi (P < 0.01). Mezenter dokusunun histamin düzeyleri ise çok önemli farklılık gösterdi (P<0.001) ve ortalama değerleri kontrollerde 20.8552 y/ gr, E vitaminlilerde 2.1504 ±0.1156 y/gr di. Histolojik değerlendirmede kontrol grubu preparatlannda mast hücre sayısında ve granulasyonunda artış, aynca hücre-hücre birleşmeleri gözlendi. E vitaminli farelerde ise çok az sayıda soluk renkte boyanmış hücrelere rastlandı. Bu çalışmadaki histamin düzeyleri ile histolojik değişiklikler bir birini desteklemektedir ve bu bulgular E vitaminin mast hücre hiperaktivitesini stabilize edici etkisi olduğunu göstermektedir. | |
dc.description.abstract | -28- VII. SUMMARY In this study, the relationship between mast cells and vitamin E was examined. Fourteen mice which had been immunized, were used in the expe riment. Animals were seperated in to two groups, one of them was control group, other was experiment group. During ten days, i.m. 0,002 ml/gr SF was injected to control group (7), i.m. 0,3 mg/gr alpha tocoferol asetate (vitamin E) was injected to each of the experiment group (7). At the end of this study, histamine levels we re determined in the mesenter tissue, periton and blood samples. Moreover in the periton liquid and mesenter tissue samples, mast cells were stained with toluidin blue and examined under light microscope. While the levels of histamine in blood of control group was 1.6781±0.4465 y/ml, it was 0.238710.0543 y/ml in experiment group and the dif ference between the two groups were found significant (P<0.01). While the levels of histamine in periton liquid was 1.830610.3539 y/ml, it was 0.28671 0.0964 y/ ml in experiment group, the difference between them was also found significant (P< 0.01). There was statistically significant difference in the levels of histamine in mesenter tissue (P< 0.001) and its average was 20.8552 y/gr in control group and 2.150410,1156 y/gr in experiment group. Histologically, the increase of num ber and granulation of mast cell were seen in preparates of control group, moreo ver cell-cell combination was seen in them. And very little amount of pale colou red cells were found on mice which had been given vitamin E. In this study the levels of histamine and histological differences we re found to support each other and these results show that vitamin E has an effect of stabilizing the hiperactivity of mast cells. | en_US |
