dc.description.abstractTEZ KONUSU:Koroner kalp hastası bireylerde serum Paraoxonase/Arylesterase aktivitesi ile serum lipidleri arasındaki ilişkinin incelenmesi Ö?RENCİMİN ADI SOYADI:Sündüs İbiş TEZ DANIŞMANININ ADI SOYADI:Prof. Dr. A.Esat Karakaya ANADİLİM DALI:Farmasötik Toksikoloji YILI:1994 STATÜ:Yüksek Lisans ÖZET Bu çalışmada paraoksonazın ilpid metabolizmasında görevli bir enzim olabileceği ve bu nedenle koroner kalp hastalığı ile aralarında bir ilişki kurulabileceği düşüncesiyle koroner kalp hastası (KKH) bireylerde serum paraoksonaz aktivite¬ si ile serum lipid (total kolesterol, trigliserld ve HDL- kolesterol) düzeyleri incelenmiştir. Yapılan ölçümler sonucunda KKH toplam bireylerin paraoksonaz akitviteleri kontrol grubundan anlamlı farklı bulunmamasına karşın erkekler arasında anlamlı bir farklılık olduğu gözlenmiştir. KKH grubu erkeklerde paraoksonaz akü- vltesi kontrol grubundan daha düşük düzeyde bulunmuştur. Benzer bir ilişki kadınlar arasında tesbit edilmemiştir. Serum HDL-kolesterol ve trigliserld düzeyleri de KKH grupta kontrol grubundan farklı bulunmuştur. HDL-kolesterol kontrolden daha düşük; trigliserid düzeyi ise daha yüksek bulunmuştur. Paraoksonaz polimorfizm gösteren bir enzimdir. Çalışmamızda da kontrol ve KKH gruplarında paraoksonaz polimorfizmi tesbit edilmiştir. Her iki grupta da trimodal dağılım görülmüş ancak fenotlpler arası dağılımda anlamlı bir farklılık bulunmamıştır. Fakat KKH grupta A fenoüpi olma olasılığı yüksek bulunmuştur. It is shown that there is an inverse relation between high-density-lipoprotein-cholesterol (HDL-C) and the risk of coronary heart disease, and paraoxonase may play a role in lipid metabolism and preventing of accumulation of the lipoperoxides. Becauce of these, we investigated a possible relationship between CHD and paraoxonase activity. In the measurements, although all patient's serum paraoxonase activities were not found different from controls, men patient's enzyme activities were found lower than the corresponding group of control. The difference was statistically significant. But the same relation was not found between women.

Patient's HDL-C values were lower and triglycerides were higher than controls. We found that the activity of human serum paraoxonase exhibited a genetic polymorphism in both control and CHD populations as had been shown previously. Paraoxonase activity was trimodally distributed in both two groups but there was not found any statistically significant differences in phenotype distributions between control and CHD patients. However in CHD group, a high possibility was found to be phenotype A compaired with the control group.en_US
