Bu çalışmada sar; boyalarına mesleksel olarak maruz kalan kuaförlerin idrar muta jeni t.eier i araştırılmıştır. Ankara'da çalışan yaşları 17-43 arasında değişen, Pazar günü hariç her gün, 2-25 yıl süreyle, günde 2-10 sefer eldivensiz olarak sac boyası uygulayan kuaförden çalışma günü sonundaki ve sabahki idrarları toplanmıştır. îörar örnekleri XAD-2 reçinesi kullanılarak konsantre edildikten sonra DMSO ile resüspanse edilmiştir. Düzeltilmiş muta jenik aktiviteyi hesaplamak amacıyla idrar örneklerinin kreatinin değerleri ölçülmüştür. Her idrar örneği Salmonella typhimur ium TA98 susunu kullanarak, rni krozoma 1 enzim fraksiyonlu (+S9) ve fraksiyon ilavesi olmaksızın (-S9) ve fî-glukuroni daz varlığında Arnes teste uygulanmıştır../ :' `` ı<.?7Tt?ı !WV':activity were observed due to occupational exposure. dc.description.abstractSUBJECT ofthe THESIS : Evaluation of Mutagenic Activity in HairDye Appliers NAME of the STUDENT: Neslihan Aygün NAME of the SUPERVISOR: Semra Sardaş DEPARTMENT: Toxicology YEAR: 1995 STATUS: Master S U fMARY Most chemical constituents of hairdye formulations have been shown to be mutagenic/carci nogeni c in bacterial strains and rodents. In this study urine mutagenecity of hair-dressers who are occupat ional ly exposed to hair dyes were investigated. Urine samples at the end of the working day and morning were collected from hair-dressers working in Ankara everday except Sundays (age ranging between 17-43, who applies hair-dye to their customers 2-10 times/daily without wearing gloves for 2-25 years). Urine samples were concentrated by using XAD-2 recine and resuspended in DMSO. Creat. inin values of urine samples were measured in order to evaluate the corrected mutagenic activity. Every single sample were applied to Ames test by using Samonella typhimurium TA 98 strain, microsomal enzyme fraction (+S9) and without -S9 plus (2-g lucoroni dase. Revertant colony counts obtained from 15 exposed unprotected hairdressers CS9 (+ ) 3. 80+8. 23, S9 (-) 1. 5±2. 261 were compared by the results of 10 control subjects CS9(+> 1.67+0.98, S9 (-) 1. 24±0. 81 1 by using the correctedactivity were observed due to occupational exposure.
