dc.description.abstract8-hidroksi-2'-deoksiguanozin (8-OHdG) DNA lezyonlannda en 90ktespit edilen ve çahsilan iiriindiir. Okside DNA, tamir sistemleri ile kendinionanrken okside bazlar idrar ile atihrlar. idrar 8-OHdG diizeyleri, kanser gibidejeneratif hastahklara bagh hiicresel oksidatif stresi ölçmek için iyi birbelirteçtir.Qahsmamizda mesane, prostat ve böbrek hiicreli karsinomdan(BHK) olusan hastalann ameliyat öncesi ve sonrasi idrar 8-OHdG diizeyleriniölçerek tedavi etkinligini görmeyi amaçladik. Ayrica saghkh kontrol, benignprostat hiperplazisi (BPH) olan hastalar ile tas rahatsizhgi olan kisileri dekarsilastirma için çahsmaya dahil edildi. Hastalardan toplanan idrarlar yiiksekperformansh SIVI kromatografisi ve elektrokimyasal dedektör (HPLC-EC) ileçahsildi.Kanser hastalanndan (mesane, prostat ve böbrek hiicrelikarsinom) olusan grup I'in ortalama 8-OHdG/kreatinin seviyeleri (0,143 ± 0,097)sirasiyla kontrol, BPH ve tas hastalanndan daha yiiksek bulunmustur (0,084 ±0,036, 0,067 ± 0,017, 0,067 ± 0,029, p<0,05). Kontrol grubu ile BPH ve tashastalan arasinda ise bir fark bulunamamistir. Ayrica mesane ve prostatkanserli hastalannda 8-OHdG/kreatinin seviyeleri kontrol grubuna göre dahayiiksektir (p<0.05). Ameliyat olan 14 hastadan 12 tanesinde (%85,7) 8-OHdG/kreatinin azalmistir ve bu diisiis istatistiki olarak anlamhdir (0,135 ±0,093 & 0,092 ± 0,034, p<0,05).Bizim bulgulanmiza göre 8-OHdG/kreatinin degerleri, kanserhastalannda olusan oksidatif DNA hasanni ölçebilecek potansiyel birbelirteçtir. Ayrica cerrahi tedavi sonrasinda bakilacak 8-OHdG/kreatininseviyeleri tedavinin ne kadar etkili oldugunu gösterebilir.
dc.description.abstract8-hydroxy-2?-deoxyguanosine (8-OHdG) is the most frequentlydetected and studied DNA lesion. Upon DNA repair, 8-OHdG is excreted in theurine. Urinary 8-OHdG is now considered as a biomarker of generalized,cellular oxidative stress and is linked to degenerative diseases includingcancer.In this study it is aimed to analyze efficiency of assaying urinaryexcretion of 8-hydroxy-2?-deoxyguanosine in patients undergoing surgery forurological cancer (bladder, prostate and renal cell cancer (RCC)). We have alsoassay groups including benign prostatic hyperplasia (BPH), healthy controland the patients with renal stone. Urinary samples were studied with highperformanceliquid chromatography and electrochemical detector (HPLC-EC).Group 1 included cancer patients (bladder, prostate and RCC) withan average of 8-OHdG/creatinine rate (0,143 ± 0,097) is sequentially found to behigher than control, BPH and renal stone groups (0,084 ± 0,036, 0,067 ± 0,017,0,067 ± 0,029, p<0,05). On the other hand, no significant difference is observedwithin control group, BPH and renal stone patients. Furthermore, bladder andprostate cancer patients? 8-OHdG/creatinine rates has found to be higher thancontrol group (p<0,05). Within 14 patients undergoing surgery 12 (85,7%) haddecreasing values of 8-OHdG/creatinine rates, indicating a statisticalsignificance (0,135 ± 0,093 & 0,092 ± 0,034, p<0,05).As a result of our findings, it is concluded that 8-OHdG/creatininerates are potential biomarkers for detecting oxidative DNA damage in cancerpatients. 8-OHdG/creatinine rates can also be used as an indicator of surgicaltherapy.en_US
