Epstein-Barr viruse (EBV), başta enfeksiyöz mononükleoz (EM) olmak üzere Burkit lenfoma ve nazofarengeal karsinoma gibi malignitelerin ayrıca transplantasyon sonrası immünsüpresif konakta ortaya çıkan posttransplant lenfoproliferatif hastalığın (PTLH) etiyolojisinden sorumlu, tüm dünyada erişkinlerin %90'ını enfekte eden insan herpesvirüs ailesinin bir üyesidir. Yaptğımız çalışmda 60 hasta serum örneğinin spesifik EBV antikorlarına VCA-IgG, VCA IgM, EBNA antikorları ve EA IgG antikoru (Diagnostic Bioprobes Srl-Italy) kiti kullanarak ELISA tekniği, EBV DNA'sına gerçek zamanlı PZR tekniği ile (artus EBV QS-RGQ Kit (24) CE-Qiagen-Almanya) kiti kullanrak bakıldı. Çalışmamızda EBV DNA'sı pozitif bulunan 17(%28.33) hastadan 4(%23.5) hasta öneğinin 3(%17.64)'ü çocuk hematoloji-onkoloji bölümünden 1(%5.88)'si kemik iliği transplantasyon ünitesinden gelen hastalar seronegatif olarak saptanmıştır. Çalışmamızda serolojik olarak EBV enfeksiyonu düşünülen hastalarda gerçek zamanlı PZR yönteminin EBV enfeksiyonunun tanı ve evrelendirmedeki öneminin belirlemesini amaçlanmıştır.Anahtar kelimeler: Epstein Barr Virüs, moleküler, serolojik, ELISA, PZR
Epstein-Barr viruse (EBV) is a member of human herpesvirus family seen in particularly infectious mononucleosis (EM), including malignancies such as Burkitt 's lymphoma and nasopharyngeal carcinoma, as well as responsible for the etiology of posttransplant lymphoproliferative disease emerging in immunosuppressive host (PTLH) after transplantation and that infects more than 90% of the adults. In our study, serum samples of 60 patients have been observed for their specific EBV antibodies of VCA-IgG, VCA-IgM, EBNA antibody and EA IgG by using (Diagnostic Bioprobes Srl-Italy) kit with the ELISA technique, EBV DNA has been observed by real-time PCR technique by using (artus EBV Kit QS-RGQ (24) CE-Qiagen, Germany) kit. In our study, of 17 patients which were found positive in EBV DNA (%28.33), 4(%23.5) of patient samples, three samples (17.64%) were children's hematology-oncology section, 1 (5.88%) was found to be seronegative patients from the bone marrow transplant unit. In our study, the purpose is to determine the importance of PZR real-time method fort he diagnosis and staging in patients with suspected serological EBV infection. Keywords: Epstein Barr Virus, molecular, serological, ELISA, PCRen_US
