dc.description.abstractİSTAVRİT {Trachurus trachurus) BALIĞINDAN İZOLE EDİLEN LAKTİK ASİT BAKTERİLERİNİN METABOLİK VE ANTİMİKROBİYAL AKTİVİTELERİNİN İNCELENMESİ (Yüksek Lisans Tezi) Fatma İLYASLI GAZİ ÜNİVERSİTESİ FEN BİLİMLERİ ENSTİTÜSÜ Mart 1997 ÖZ Araştırmada, istavrit balığı {Trachurus trachurus) örneklerinden toplam 16 adet Lactobacillus izole edilmiştir. Lactobacillus 'lann tamamının Betabacterium IIA ve IIB grubuna dahil olduğu tespit edilmiştir. Bakterilerin idendifıkasyon sonuçlan 10 adet Lactobacillus hilgardii, 3 adet Lactobacillus confusus, birer adet Lactobacillus cellobiosus, Lactobacillus fermentum ve Lactobacillus fructivorans olduğunu göstermiştir. Bakterilerin ürettiği laktik asit miktarı %10 'hık yağsız süt tozu besiyerinde titrasyon metodu ile, hidrojen peroksit MRS sıvı besiyerinde kantitatif olarak spektrofotometrik yöntemle saptanmıştır. Lactobacillus suşlarının %0. 18-0. 108 oranında laktik asit, 1.74-4.46 j.g/ml arasında hidrojen peroksit ürettikleri saptanmıştır. Suşların hidrojen sülfür üretimleri Triple Sugar Iron Agar (TSİ) 'da belirlenmiştir. Suşların hiçbirisi hidrojen sülfür üretmemiştir. Suşlann proteolitik aktivite değeri 0.002-0.125 mg tirosin/ml arasında bulunmuştur. Lactobacillus suşlarının Escherichia coli, Staphylococcus aureus koagülaz (+), mutant Staphylococcus aureus koagülaz (-), Pseudomonas aeruginosa, Enterobacter aerogenes ve Pseudomonas fluorescens üzerindeki inhibisyon etkileri agar diffuzyon yöntemiyle belirlenmiştir. Bu bakterilerin bakteriosin üretimleri, aynı test bakteriler ile Micrococcus flaws üzerinde uygulanmıştır.n Lactobacillus türlerinin E. coli üzerindeki genel inhibisyon etkileri 0.00-9.20 mm. arasında, S. aureus koag.(+) 4.20-12.60 mm, S. aureus koag.(-) 7.30-15.60, P. aeruginosa 4.50-9.70 mm, E. aerogenes 1 1.00-13.40 mm ve P. fluorescens 8.40-1 1.40 mm yarıçapında genel inhibisyon etkileri gözlenmiştir. L. cellobiosus ve L. fructivorans suşları E. coli üzerinde, L. fermentum ve L. cellobiosus S. aureus coag.(-) üzerinde herhangi bir inhibisyon göstermemiştir. Bakterilerin S. aureus koag.(+) üzerindeki bakteriosine bağlı inhibisyon etkileri 2.00-8.70 mm, S. aureus koag.(-) 3.30-8.00 mm, M flavus 1.50-9.00 mm olarak gözlenmiştir. Lactobacillus suşlarının, %2-%20 arasında NaCl içeren MRS besi ortamında üremeleri incelenmiştir. Sonuçlar, 15 Lactobacillus susunun %20 NaCl 'de üreme yeteneğine sahip olduklarım göstermiştir. dc.description.abstractIll METHABOLIC AND ANTIMICROBIAL ACTIVITIES OF LACTIC ACID BACTERIA ISOLATED FROM {Trachurus trachurus) FISHES (M.Sc. Thesis) Fatma İL YASLI GAZI UNIVERSITY INSTITUTE OF SCIENCE AND TECHNOLOGY March 1997 ABSTRACT Sixteen Lactobacilli were isolated from Trachurus trachurus fishes. Identification of the strains of Lactobacilli showed that they were belong to heterofermantative Betabacterium IIA and IIB. Only 10 strains were identified as Lactobacillus hilgardii, 3 strains were identified as L. confusus, 1 strain was identified as L. cellobiosus, 1 strain was identified as L. fructivorans, 1 strain was identified as L. fermentum. Strains were inoculated to 10 % skim milk medium, and the amount of lactic acid produced by the strains were estimated by titration method. Amount of H2O2 produced by the strains were estimated in MRS Broth medium by spectrophotometric method. Amount of lactic acid and H2O2 produced by Lactobacillus strains were ranged between 0.18-0.108 % and 1.74-4.46 jig/ml, respectively. A qualitative method, Triple Sugar Iron Agar, were used to estimate hydrogen sulfide production by Lactobacillus strains. All of Lactobacilli strains did not produce hydrogen sulfide. Proteolytic activity of the Lactobacillus strains were measured between 0.002-0.125 mg of tyrosine/ml. Agar diffusion method was used to study for the bacterial inhibition on Escherichia coli, Staphylococcus aureus coagulase (+), mutant S. aureus coagulase (-), Pseudomonas fluorescens, P. aeruginosa andrv Enterobacter aerogenes. Inhibition was calculated by measuring the half diameter of inhibition zones of the tested microorganisms. Lactobacillus strains inhibited E. coli by inhibition zones of 0.00-9.20 mm, S. aureus coag.(+) 4.20-12.60 mm, S. aureus coag.(-) 7.30-15.60 mm, P. aeruginosa 4.50-9.70 mm, E. aerogenes 1 1.00-13.40 mm and P. fluorescens 8.40-1 1.40 mm of the half diameter of inhibition zones. Any inhibition was not seen for L. cellobiosus and L. fructivorans on E. coli and, for L. cellobiosus and L, fermentum on S. aureus coag (-). Bacteriocin-dependent inhibition by Lactobacillus strains was tested on the test bacteria and Micrococcus flavus. Bacteriocin-dependent inhibition zones for S. aureus coag.(+), S. aureus coag.(-) and M. flavus were in 2.00-8.00, 3.30-8.00 and 1.50-9.00 mm of the half diameter, respectively. Other test bacteria were not inhibited by bacteriocin produced by Lactobacillus strains. Growth of Lactobacillus strains in MRS Broth medium containing 2-20 % NaCl was also studied. Results showed that the 15 Lactobacillus strains were capable to growth in 20% of NaCl concentration.
