Abstract
SUMMARY In this study, the effects of antifungal, antineoplastic and antipsychotic drugs alone and in combination were investigated on standard and clinical strains of Candida albicans. Sabouraud broth was preferred to YNB+glucose broth to determine the MIC values. Among the three methods, namely, adjusting the overnight culture to 90% transmittance at 530 nm, or to 0.200 absorbance at 600 nm or diluting the overnight culture 10 times, The inoculum obtained by 10 dilution of an overnight culture was used in the studies since it gave reproducible results that were parallel to those in the literature. MIC values ranging between 15.625 and 250 ug/ml were obtained with antineoplastic and antipsychotic drugs against standard and clinical strains of C. albicans. 51In combination studies synergy, in the combinations of amphotericin B and ketoconazol with tamoxifen bieocin metotho- trexate and trifluoperazine was observed. Interactions of these drugs with nystatin showed antagonistic, additive and indiffe rent effects. The postantifungal effect was quantitaded by hourly viable counts of cfu's after removal of the drugs by dilution technique. The longest postantifungal effect were obtained with amphotericin B (`5 h) and the lowest with tamoxifen {`0.18 h). There was no significant difference between PAE's obtained with standard and clinical strains of C. albicans. There wasn't a significant increase in the PAE values of drug combinations against standard and clinical strains of C. albicans. Meanwhile, PAE values obtained by drug combinations with amphotericin B and nystatin were lower then the values when the antifungals were used alone.
