Kudret Narı` nın (Momordica charantia) östrojen reseptörlerinden ESR1 ve ESR2 gen düzeyleri ile oksidatif DNA hasarı üzerindeki etkisi

Abstract
Menopoz döneminde ortaya çıkan değişiklikler, bayanların yaşam kalitesinin düşmesine ve çeşitli hastalıkların görülmesine yol açmaktadır. Bu değişikliklerin temel nedeni dolaşım sisteminden 17-ß Östradiol (E2) hormon düzeyinin azalması ve yerini östron hormonuna bırakmasıdır. Son yıllarda fitoöstrojen içeriği yüksek olan bitkiler menopoz döneminde tedavi edici olarak tavsiye edilmektedir. Bu çalışmada deneysel overektomize modelinde, Momordica charantia olarak isimlendirilen Kudret Narının östrojen reseptörlerinden ESR1 ve ESR2 gen düzeyleri ile oksidatif DNA hasarı üzerindeki etkisini araştırmayı amaçladık. Çalışmada Wistar albino türüne ait sıçanlar ligatürasyon yöntemi ile overektomi modeli oluşturularak 2 gr/kg Kudret narı meyve ekstresi (KN) gavaj ile 30 gün boyunca uygulandı. Serumda E2 ve 8-OHdG düzeyleri ile uterus, karaciğer ve böbrek dokusunda MDA, GSH, SOD, GST, CAT ve MPO düzeyleri ölçüldü. ESR-1 ve ESR-2 östrojen genleri ve NF-kB düzeyleri RT-PCR ile uterus dokusunda araştırıldı. Uterus, karaciğer ve böbrek dokusundaki yapısal değişiklikler HE boyaması ile ışık mikroskobunda incelendi. Bulgularımızda overektomize sıçanlara uygulanan KN serum E2 düzeyini arttırmış, 8-OHdG düzeyini azaltmıştır. Overektomide KN uygulaması ile ESR-1, ESR-2 ve NF-kB gen düzeyleri anlamlı olarak artmıştır. Karaciğer, böbrek ve uterus dokusunda overektomi sonucunda değişiklik gösteren oksidatif strese karşı KN uygulamasının antioksidan etki gösterdiği bulunmuştur. Histolojik bulgular ele alındığında özellikle uterus dokusunda overektomide KN uygulamasının nötrofil infiltrasyonunu azalttığı, hücre ve kollajen dağılımının normalle döndüğü, düzgün yapıda epitel hücrelerinin arttığı tespit edilmiştir. Momordica charantia bitkisi fitoöstrojen ve antioksidan etkisi ile overektomili sıçanlarda hem gen düzeyinde hem de protein düzeyinde etki göstermiş olup menopoz döneminde kullanılabilecek yeni ilaçların geliştirilmesinde katkıda bulunabilir.Anahtar Kelimeler: Momordica charantia, Overektomi, 17-ß Östradiol, Östrojen reseptörleri, Oksidatif DNA Hasarı Changes arising in the period of menopause, cause the quality of life to reduce various diseases to appear. Main reason of these changes, is the decrease of 17-ß estradiol (E2) hormone level in the circulation system and estrogen hormone taking its place. In recent years plants with high phytoestrogen content are recommended as therapeutically. In this study, we aimed at investigating the effect of Bitter Melon (BM) named as Momordica charantia on the gene levels of estrogen receptors such as ESR-1 and ESR-2, and on oxidative DNA damage, in experimental ovariectomized model. During the study, the ovariectomy model was composed by ligaturation method and 2g/kg of fruit extra gavage of Bitter Melon applied on the rats belonging to the species of Wistar albino, for 30 days. Inside the serum, E2 and 8-OHdG levels; MDA, GSH, SOD, GST, CAT and MPO levels in uterus, liver and kidney tissue, were measured. Estrogen genes ESR-1 and ESR-2 and NF-kB along with RT-PCR, were analyzed in uterus tissue. Structural changes in uterus, liver and kidney tissue, were examined on the light microscope by means of HE staining. In our findings, BM serum applied on ovariectomized rats has increases its E2 level, and decreased 8-OHdG level. In ovariectomy, ESR-1, ESR-2 and NF-kB gene levels were increased significantly by BM application. It was discovered that BM application had reacted with an antioxidant effect against the oxidative stress that showed an alteration in liver, kidney and uterus tissue as a result of the ovariectomy. When histological findings were taken into consideration, it was observed that; BM application had decreased its neutrophils infiltration in the ovariectomy especially in the uterus tissue, cell and collagen distribution had returned to normal, and epithelium cells had increased in the uniform structure. Momordica charantia plant displayed an effect on ovariectomized rats in both gene and protein level, by the effect of phytoestrogen and antioxidant; and it may contribute to the development of new medicine that can be used in the period of menopause.
