Abstract
Amaç: Bu çalışmada MCF-7 meme kanseri hücre soylarında diosgeninin maddesinin anti-kanserojen etkisinin, PI3K/Akt ve mTOR inhibitörü olan daktolisib ile nasıl değiştiğini belirlenmesi amaçlandı. Çalışmamızda, meme kanseri MCF-7 hücre hattında diosgenin, daktolisib ve onların farklı kombinasyonlarının kullanılmasıyla bu maddelerin meme kanseri hücreleri üzerindeki sitotoksik ve apoptotik etkileri belirlendi.Gereç ve Yöntem: Diosgenin ve daktolisibin büyüme inhibisyonu, MCF-7 hücre soylarına karşı MTT kalorimetrik yöntem kullanılarak in vitro olarak değerlendirildi. Farklı konsantrasyonlarda diosgenin ve daktolisibin MCF-7 meme kanseri hücre soyları üzerindeki apoptotik etkileri Tali görüntü tabanlı sitometre ve mitokondriyal membran potansiyeli değişim testi kullanılarak belirlendi. ¼ IC50, ½ IC50 ve IC50 değerlerinde diosgenin ve daktolisibin reaktif oksijen türleri deneyi yapıldı. Ayrıca Calcusyn programı kullanılarak iki ilacın kombinasyon deneyleri için uygun dozlar saptandı. Bulgular: Diosgenin ve daktolisibin %50 inhibisyon konsantrasyonu sırasıyla 85 M ve 20 nM'dır. Ayrıca diosgenin ve daktolisib, MCF-7 hücrelerine ayrı ayrı uygulandığında artmış apoptotik etkileri indüklediği bulundu. İki ajanın antagonist etki gösterdiği belirlendi. Sonuç: Diosgenin ve daktolisibin belirlenen konsantrasyonlarda kullanımının meme kanseri tedavisi için önemli olabileceğini, fakat iki ilacın kombine kullanımının meme kanseri tedavisine etkisinin olmadığını düşünmekteyiz.Anahtar Kelimeler: Diosgenin, daktolisib, MCF-7, meme kanseri, apoptoz. Aim: In this study, we aimed to determine how anti-carcinogenic effect of diosgenin agent in MCF-7 breast cancer cell lines changes with daktolisib, PI3K / Akt and mTOR inhibitor. In our study, the cytotoxic and apoptotic effects of these substances on breast cancer cells were determined by using diosgenin, dactolisib and their different combinations in the breast cancer MCF-7 cell line.Methods: Growth inhibition of diosgenin and dactolis was evaluated in vitro using the MTT calorimetric method against MCF-7 cell lines. Apoptotic effects of diosgenin and dactolisib in different concentrations on MCF-7 breast cancer cell lines were determined by using Tali image based cytometer and mitochondrial membrane potential change test. ¼ IC50, ½ IC50 and IC50 values of diosgenin and Dactolisibin reactive oxygen species were tested. Also, the appropriate doses were determined for the combination experiments of the two drugs using the Calcusyn program.Results: The 50% inhibition concentration of diosgenin and dactolisib was 85 M and 20 nM, respectively. Also diosgenin and daktolisib were found to induce increased apoptotic effects when administered separately to MCF-7 cells. Two agents showed antagonistic effects.Conclusion: We believe that the use of diosgenin and dactolis in determined concentrations may be important for the treatment of breast cancer, but the combined use of the two drugs has no effect on breast cancer treatment.Keywords: Diosgenin, dactolisib, MCF-7, breast cancer, apoptosis.
