Abstract
Bu çalışmada füloresan Nil kırmızısı probu kullanılarak üretici mikroorganizmaların polihidroksialkanoat üretim kapasiteleri izlenmiştir. Öncelikle kullanılan dört farklı mikroorganizma olan Ralstonia eutropha, Aîcaligenes lotus, Pseudomonas aeruginosa ve Escherichia colV nin üreme parametrelerini inceleyen ayrıntılı bir çalışma yapılmıştır.Daha sonra üretici suşlann polimer üretimi sırasındaki davranışları incelenmiş ve bu polimerlerin hücrelerden elde edilmesi için gerekli şartlar optimize edilmiştir.Nil kırmızısı uygulanan hücreler mikroskopik olarak incelenmiş ve spektroflorometre ile üretilen polimer miktarım Nil kırmızısı boyaması sonucunda oluşan floresans ile bağlayan kalibrasyon eğrileri çizilmiştir. Bu yöntem ile hücre içindeki polimer konsantrasyonu hücre parçalanmadan belirlenebilmektedir. Bu sonuçlara bağlı olarak doğadaki potansiyel polimer üreticilerini belirleyebilmek için bir koloni tarama metodu da geliştirilmiştir. Anahtar kelimeler : Nil kırmızısı, Polihidroksialkanoatlar, Ralstonia eutropha, Aîcaligenes latus, MTT, İzleme IV In this study, polyhydroxyalkanoate production capacities of producer microorganisms were monitored by using a newly established method employing fluorescent Nile red as a probe. A detailed study, involving growth parameters was performed first with four strains of Ralstonia eutropha, Alcaligenes latus, Pseudomonas aeruginosa and Escherichia coll Next the behaviour of the strains during the polymer production was followed and extraction of these polymers from cells were optimised. Nile red applied cells were examined microscopically and spectrofluorimetrically leading to the construction of a calibration curve which linked the concentration of the polymer present to the florescence intensity after Nile red staining. By this way the polymer production could be quantified within the cells without destruption. A colony screening method, based on these findings, was also developed to monitor potential polymer producing strains in nature.
