Abstract
ÖZET Tip 2 diabetes mellitus'ta etkin rolü olan apolipoprotein E ( apo E ) plazma lipoprotein metabolizması düzenleyicilerindendir. Apo E geni polimorfiktir ve sık rastlanan 3 alleli ( e2, e3, s4 ), apo E2, E3, E4 izoformlannı kodlar. Çalışma tedavi görmekte olan 126 diabetik (45 erkek, 81 kadın) ve 107 kontrol (29 erkek, 78 kadm) örnekleri üzerinde yapıldı. Apo E polimorfizmleri, izole edilen DNA örneklerinden polimeraz zincir reaksiyonu ve Hhal restriksiyon izotiplemesi ile tespit edildi ve apo E genotiplerinin serum lipid ve apolipoproteinlerinin düzeyleri üzerine olan etkileri incelendi. Tip 2 diabetik grupta allel sıklığı apo E2, E3, E4 için sırası ile 0.07, 0.846, 0.083, kontrol grubunda ise 0.076, 0.814 ve 0.109 olarak saptandı. Diabetin serum lipid profili üzerine olan etkileri cinsiyete göre farklı bulundu. Serum kolesterol, apolipoprotein A-I ve apo E konsantrasyonları her iki grupta kadınlarda yüksek bulunurken ( p<0.01), lipoprotein (a) düzeyleri erkeklerde daha yüksek tespit edildi. Apo E2 allel sıklığı cinsiyetler arasında farklı olup, kadınlarda erkeklerin iki katı olarak tespit edildi. Apo E allelleri arasında apo A-I (p<0.05) ve apo E (p<0.001) düzeyleri farklılık göstermekte idi. Diabetik grupta en yüksek apo A-I ve apo E düzeyleri apo E2 genotipine sahip kişilerde görüldü. Trigliserid düzeylerinin erkek diabetik ve kontrollerde apo E allel dağılımlarına bağlı olarak değişiklik gösterdiği saptandı (p<0.05). 58 SUMMARY Apolipoprotein E ( apo E ) is one of the organisers of plasma lipoprotein metabolism which has a major role in type 2 diabetes mellitus. Apo E gene is polymorphic and its 3 allele ( s2, e3, e4 ), respectively codes for apo E2, E3, E4 isoforms. The study was performed on 126 diabetic ( 45 men, 81 women ) and 107 control ( 29 men, 78 women ). After DNA isolation from the samples, apo E polymorphisms were detected with polymerase chain reaction and Hhal restriction isotyping. The effect of apo E genotypes on serum lipid and apolipoprotein levels were also investigated. The relative allele frequencies of apo E gene in the diabetic group was found to be 0.07 for E2, 0.846 for E3, 0.083 for E4, and in the control group 0.076 for E2, 0.814 for E3 and 0.109 for E4. The effects of diabetes on serum lipid profile was found different with gender. Serum cholesterol, apolipoprotein A-I and apo E concentrations were found higher in women (p<0.01), whereas lipoprotein (a) levels were higher in men for both diabetics and controls. Apo E2 allele frequency was different for gender and twice in women compared to men. Apo A-I (p<0.05) and apo E (pO.OOl) levels were different with respect to apo E alleles. Apo A-I and apo E levels were detected to be the highest in individuals with apo E2 genotype. Triglyceride levels were differing with respect to apo E alleles in male diabetic and controls. 59
