Nimesulidin jel tipi farmasötik preparatlarının hazırlanması ve stabilitesi üzerine çalışmalar

Abstract
ÖZET Bu çalışmanın amacı, oral preparat şekilleri ile tedavide nonsteroidal antienflamatuar bir ilaç olarak kullanılan nimesulidin lokal jel tipi farmasötik preparatını hazırlamaktır. Jel formülasyonlartnın hazırlanmasında jelleştirici madde olarak Carbopol® 934 kullanıldı. Çalışmada jel formülasyonlarında kosolvan olarak etanol, izopropil alkol, Transcutol® ve propilen glikol, penetrasyon artırıcı olarak ise Tween 80® ve oleik asit kullanıldı. Formülasyonlarda yer alan kosolvan ve/veya penetrasyon artırıcı maddelerin etkisini değerlendirmek amacıyla kosolvan ve penetrasyon artırıcı madde içermeyen kontrol jel formülasyonu da hazırlandı. Formülasyonların pH sı 6.0±0.1 olarak sabit tutuldu. Hazırlanan bütün formülasyonlarda nimesulid miktar tayini yapıldı ve formülasyonların viskoziteleri ölçüldü. Formülasyonlardaki nimesulidin in vitro serbestleşme deneylerinde selofan zar ve modifiye Franz difüzyon hücresi kullanıldı. Belirli zaman aralıklarında 1 cm2 lik alandan serbestleşen nimesulid miktarları hesaplanarak nimesulidin in vitro serbestleşme hızı profilleri çizildi ve formülasyonlar için nimesulidin serbestleşme hız sabiteleri hesaplandı. Elde edilen bulgular kinetik modellere uygulanarak değerlendirildi. Kontrol jel formülasyonu ile karşılaştırılarak seçilen en uygun formülasyonlardaki nimesulidin ex vivo sıçan derisinden difüzyonu incelendi. Ex vivo deneylerde sıçan karın derisi kullanıldı. Zamana karşı formülasyonlardaki nimesulidin 1 cm2 lik alandan difüze olan miktarlarına ait difüzyon profilleri çizildi. Nimesulidin sıçan derisinden geçiş hızı, gecikme süresi ve permeabilite katsayısı üzerine formülasyonlardaki kosolvan ve/veya penetrasyon artırıcı maddelerin etkisi, kontrol formülasyonu ile karşılaştırmalı olarak değerlendirildi. Ayrıca bu formülasyonlarda nimesulidin 3 ay süre ile 25±2°C, %60±5 RH; 30±2°C, %60±5 RH; ve 40±2°C, %75±5 RH koşullarındaki stabilitesi izlendi. Elde edilen ex vivo deney bulgularının istatistiksel olarak değerlendirilmesi ile, %40 izopropil alkol içeren F4 ve %3 oleik asit - %30 Transcutol® içeren F10 formülasyonlarının, kontrol formülasyonuna göre nimesulidin sıçan derisinden geçiş hızı ve permeabilite katsayısını anlamlı 100olarak artırdığı saptandı. Ancak oleik asit içeren formülasyonda (F10) ilacın deriden geçişi için gecikme süresinin uzun olduğu (2.19 saat), izopropil alkol içeren formülasyon (F4) için bu sürenin kontrol jel formülasyonuna göre oldukça kısa olduğu (0.33 saat) saptandı. Ayrıca F4 formülasyonunun da F10 a göre nimesulidin sıçan derisinden geçiş hızı ve permeabilite katsayısını anlamlı artırdığı ve gecikme süresini de oldukça anlamlı kısalttığı saptandı. Nimesulidin uygun fizikokimyasal özellikleri ve yüksek lipofilik karakteri (log K>2) nedeniyle lokal uygulanan bir farmasötik preparatının hazırlanmasının mümkün olabileceği gösterilmiştir. Nimesulidin deriden geçiş hızını artırması ve ilacın deriden geçişinde görülen gecikme süresini kısaltmasından dolayı, jel formülasyonlarının hazırlanmasında önerilecek en iyi formülasyonun %40 izopropil alkol içeren F4 olduğu sonucuna varılmıştır. 101 SUMMARY The aim of this study is to prepare a gel type topical dosage form of nimesulide, an orally administered non-steroidal anti-inflammatory drug, for local applications. Carbopol® 934 was used as a gelling agent. In this study, ethanol, isopropyl alcohol, Transcutol®, and propylene glycol were used as co-solvents; Tween 80® and oleic acid as penetration enhancers. In order to evaluate the effect of penetration enhancers and/or co-solvents in these formulations, control gel formulations were prepared without any co-solvent and penetration enhancer. The pH values of the formulations were kept constant at 6.0 ±0.1. Nimesulide assay was performed for all the formulations and viscosity values were determined for all the gel preparations. For in vitro release studies, cellophane was used as the membrane and gels were applied onto modified Franz diffusion cells. At certain time intervals, the amount of the drug released from 1 cm2 area of the membrane was calculated and then, the in vitro release profiles of nimesulide were plotted against time. From these plots the release rate of nimesulide from the formulations were calculated and the data were kinetically evaluated. These data were compared to those obtained for control gel formulations and then, the most suitable formulations were selected. The ex vivo diffusion behavior of nimesulide from rat skin was investigated for these formulations. In these experiments, rat abdominal skin was used. The diffusion profiles for the amount of nimesulide released from 1 cm2 area of the skin were plotted against time. The effect of co-solvents and/or penetration enhancer(s) on flux of nimesulide through rat skin, lag time, and permeability coefficient was evaluated with comparison to control gel formulation. Furthermore, the stability of nimesulide in these formulations were evaluated at the conditions of 25 ± 2°C, 60% RH; 30 ± 2CC, 60% RH; 40 ± 2°C, 75% RH for three months. Based on the ex vivo data, it was determined that the formulation F4 containing 40% isopropyl alcohol and the formulation F10 prepared with 3% oleic acid-30% Transcutol® are the most efficient ones with respect to significant enhancement of flux and permeability coefficient of nimesulide 102through rat skin in comparison to those obtained for control formulations. In the formulation containing oleic acid (F10), the lag time was much longer (2.19 hours) whereas it was too short (0.33 hours) for the formulation prepared with isopropyl alcohol (F4). In this study, the possibility of topical gel formulation of nimesulide possessing proper physicochemical properties and high lipophilicity was shown. It can be concluded that the gel formulation F4, which has 40% isopropyl alcohol, is the most suitable gel preparation for topical applications of nimesulide as it enhances the flux of the active substance through the skin and shortens the lag time. 103
