xxiiÖZETPırıldar, S. (2006). Colchicum baytopiorum C.D.Brickell Üzerinde FarmakognozikAraştırmalar. İstanbul Üniversitesi Sağlık Bilimleri Enstitüsü, Farmakognozi ABD.Doktora Tezi. İstanbul.Bu çalışmada, endemik bir tür olan Colchicum baytopiorum C.D.Brickell bitkisininçiçek, çiçek sapı, yaprak, tohum ve yumruları (Haziran ve Ekim aylarında toplanan)kimyasal açıdan ve sitotoksik aktivite yönünden ayrı ayrı çalışılmış ve sonuçlarkarşılaştırmalı olarak değerlendirilmiştir.Sütun kromatografisi, kromatotron ve preparatif İTK yöntemleri uygulanarak eldeedilip UV, IR, 1H NMR ve ESI/MS spektral yöntemleri ile yapıları aydınlatılankolşisin, 2-demetilkolşisin, 3-demetilkolşisin, 3-demetil-N-formil- N-deasetilkolşisin,2-demetil-γ-lumikolşisin, kolşifolin, 2-demetilkolşifolin, demekolsin ve 2-demetildemekolsin isimli bilinen 9 adet alkaloidin varlığı saptanmıştır. Ayrıca, kornigerin, N-formil-N-deasetilkolşisin ve 3-demetildemekolsin alkaloitlerinin varlığı ise MS/MSyöntemi ile belirlenmiştir.Çiçek sapı eter ekstresinden apigenin ve luteolin isimli flavon aglikonları elde edilmiş,yapı tayininde UV ve IR spektrofotometrisi yöntemleri kullanılmıştır. Diğer eterekstreleri sadece İTK yöntemi ile karşılaştırılmış, sonuçta yaprak ekstresinde apigeninve luteolin, çiçek ekstresinde ise sadece apigenin varlığı saptanmıştır. Tohum veyumrularda flavon bileşiği bulunamamıştır.Metanol ekstrelerinde LC/MS yöntemi kullanılarak benzoik, kumarik, vanilik,sinnamik, ferulik, kafeik asitler, 2-hidroksi-6-metoksibenzoik asit, 4-hidroksi-3-metoksibenzoik asit, 2,5-dihidroksibenzoik asit, 2-hidroksibenzoik asit ve 4-hidroksibenzoikolmak üzere toplam 11 adet fenolik asidin varlığı saptanmıştır.Metanol ekstreleri sitotoksik aktivite açısından öncelikle Brine Shrimp yöntemi ileincelenmiş ve ekstrelerin tümünün aktif olduğu görülmüştür. Bu doğrultuda ekstreler,MTT yöntemi kullanılarak K562 ve HL60 lösemi hücre serilerinde kolşisin vedemekolsin şahitleri ile karşılaştırılmış ve sonuçlar Student-t testi iledeğerlendirilmiştir. Sonuçta, tohum hariç tüm ekstrelerin HL60 hücrelerine karşı K562hücrelerinde olduğundan daha sitotoksik etkide oldukları ve spektrofotometrik totalalkaloit miktar tayini sonucunda en yüksek alkaloit oranına sahip oldukları saptanançiçek (% 5,27) ve çiçek sapı (% 2,96) metanol ekstrelerinin, en yüksek aktiviteyigösterdikleri belirlenmiştir.Anahtar Kelimeler: Colchicum baytopiorum, kolşisin, tropolon alkaloitleri, fenolikasit, sitotoksik aktivite.Bu çalışma İstanbul Üniversitesi Bilimsel Araştırma Projeleri Birimi tarafındanT-435/08032004 no'lu proje olarak desteklenmiştir. xxiiiABSTRACTPirildar, S. Istanbul.In this study, flower, stem, leaf, seed and corms (collected in June and October) ofColchicum baytopiorum which is an endemic species, were studied in terms of plantchemistry and cytotoxic activity and the results were compared with each other.Nine known alkaloids namely colchicine, 2-demethylcolchicine, 3-demethylcolchicine, 3-demethyl-N-formyl-N-deacetylcolchicine, 2-demethy-γ-lumicolchicine, colchifoline, 2-demethylcolchifoline, demecolcine, 2-demethyldemecolcine were isolated and purified bychromatotron, column and preparative thin layer chromatographic methods and theirstructures were identified using spectrometric methods such as UV, IR, 1H NMR andESI/MS. Cornigerine, N-formyl-N-deacethylcolchicine and 3-demethyldemecolcine weredetected by MS/MS spectrometer.Two flavonoids, apigenine and luteoline were isolated from the ether extract of stem bypreparative thin layer chromatography and their structures were identified by UV and IRspectroscopy. These flavonoids were also found in methanolic leaf and flower extracts. Noflavonoids were found in seed and corms extracts.In all methanolic extracts 11 phenolic acids namely; benzoic, coumaric, vanillic, cinnamic,ferulic, caffeic acids, 2-hydroxy-6-methoxybenzoic acid, 4-hydroxy-3-methoxybenzoicacid, 2,5-dihydroxybenzoic acid, 2-hydroxybenzoic acid, 4-hydroxybenzoic acid weredetected using LC/MS.All methanolic extracs were subjected to cytotoxicity assay by Brine Shrimp method andall extracs were found to be active. The cytotoxicities of the extracts were evaluated usingK562 and HL60 cell lines by MTT test and all extracts were compared with colchicine anddemecolcine using Student?s t test. Except the seed extract, all extracts were found morecytotoxic on HL60 cell line than K562 cell line. The results also showed that methanolicextracts of flower (5.27 %) and stem (2.96 %), which have the richest alkaloid contents,were the most active extracts.Key Words: Colchicum baytopiorum, colchicine, tropolone alkaloids, phenolic acid,cytotoxic activity.This study was supported by The Research Support Unit of İstanbul University as theproject no T-435/08032004.
